摘 要:目的:测定紫芸解毒止痛酊中的重金属和微量元素含量,并探究其镇痛作用机制。方法:利用火焰原子吸收光谱法测得紫芸解毒止痛酊重金属含量,通过小鼠致痛模型和疼痛因子的ELASA检测初步分析镇痛作用机制。结果:紫芸解毒止痛酊不含重金属镉、铅、铬、镍,含有少量的铜和锌,浓度分别为0.0185和0.56 mg/L;紫芸解毒止痛酊能缓解急性疼痛和甲醛诱发的周围炎性疼痛,且其对皮肤的刺激性小于雪上一枝蒿速效止痛搽剂;紫芸解毒止痛酊可通过抑制前列腺素E_2并上调β内啡肽含量以达到镇痛效果,但对致痛因子一氧化氮的影响较小。结论:紫芸解毒止痛酊产品重金属含量满足药物安全性要求,为其作为外用镇痛药物的使用提供理论依据。Objective: To determine the heavy metals content of Ziyun Jiedu Zhitong Ding and preliminarily ana-lyze its analgesic mechanism. Methods: Flame atomic absorption spectrometry was used to measure the content ofheavy metal, and the pain models of mice with ELASA detection of pain factors were used to analyze mechanismof analgesic action. Results: Flame atomic absorption spectrometry showed Ziyun Jiedu Zhitong Ding did not con-tain heavy metal Ni, Pb, Cr, Cd, but contained a low amount of Cu(0.0185 μg/m L), Zn(0.56 μg/m L). Accordingto pain model data, Ziyun Jiedu Zhitong Ding alleviated the acute pain and formaldehyde induced peripheral in-flammatory pain, and the stimulus to the skin was slighter than positive drug. ELASA indicated that Ziyun JieduZhitong Ding achieved analgesia effect by inhibiting the synthesis of prostaglandin E2 and up regulation of the ex-pression of beta endorphin, however, it had a little effect on the pain factor nitric oxide. Conclusion: Ziyun JieduZhitong Ding meet drug safety regulations for heavy metals content, and the mechanism is provided for its applica-tion as the topical analgesic drug.
