摘 要:目的:研究比较中药制剂秋泻灵和儿泻停颗粒体外传代细胞上抗轮状病毒的作用,为秋泻灵的进一步动物试验及临床应用提供一定的实验依据。方法:颗粒状药物用PBS溶解过滤后作为原液,稀释至不同浓度,用于以下实验:1不同浓度的药物直接于4℃与SA11轮状病毒作用1 h后,接种到轮状病毒敏感的恒河猴肾细胞MA104上,评估药物对病毒的灭活作用;2用不同浓度的药物作用细胞1 h,再接种SA11,评估药物阻止病毒入侵细胞的作用;3先用轮状病毒SA11株感染MA104细胞,然后测定不同浓度的药物抑制病毒增殖的作用。结果:秋泻灵和儿泻停的半数细胞毒性浓度(CC50)分别为3.62和2.60 mg/m L;药物浓度为1×10^(-4)~10 mg/m L时,二者对轮状病毒有灭活作用;在阻止病毒入侵细胞的实验中,儿泻停在较高浓度时有效,而秋泻灵在低浓度时有效;在清除已感染MA104细胞的SA11轮状病毒时,二者在适宜浓度时有微弱作用。结论:秋泻灵和儿泻停在适宜浓度时对轮状病毒SA11有灭活作用;二者对阻止SA11轮状病毒入侵MA104细胞显示了一定的的保护作用;本次试验条件下,二者在一定浓度下显示了清除已感染MA104细胞的SA11轮状病毒的微弱作用。Objective: To evaluate the antiviral effect of Qiuxieling and Erxieting on SA11 rotavirus-infected Af-rican green kidney MA104 cells and to provide support to clinical treatment for diarrhea caused by rotavirus infec-tion. Methods: Cytopathic effect(CPE) of the MA104 cells caused by SA11 rotavirus was examined with MTT. Vi-ral proliferation inhibition rate of SA11 rotavirus by Qiuxieling and Erxieting, the inactive rate of SA11 and theprevention effects to SA11 were detected by MTT and evaluated. Results: The CC50 of Qiuxieling was 3.62 mg/m L and the CC50 of Erxieting was 2.60 mg/m L. The outcomes showed that there were some effects in the inac-tive viral infection of the two drugs and a little effects on preventing the SA11 rotavirus to attack MA104 cellswhen the concentration range was 1×10-410 mg/m L. The effect on clearing the viruses inside the MA104 cellswas very limited when the concentrations was appropriate. Conclusion: There are some effects on the inactivatedviral infection, inhibition rotavirus proliferation in MA104 cell lines.
