摘  要:目的对分离自明永冰川地区的1株黄杆菌噬菌体(SWP-8)生物学特性进行研究。方法利用透射电子显微镜(TEM)技术及氯化铯密度梯度离心法对SWP-8进行形态观察、基因组酶切片段长度多态性分析及噬菌体生理特性研究。结果噬菌体SWP-8为肌尾型,在电镜下呈头尾复合型结构,头部呈正多面体立体对称,直径约150nm;尾管长约150nm,直径约20nm,具有收缩性的鞘,头部与尾管结合处具有颈环结构。在4℃时具有感染性,4~25℃均能产生边缘清晰、透明的噬菌斑。pH值耐受范围较广,最适pH值为7.0。最佳感染复数为10.000,潜伏期60min,裂解期90min,裂解效率较高。结论 SWP-8属肌尾噬菌体科(Myoviridae),是裂解性噬菌体,对氯仿和蛋白酶K较为敏感。基因组为dsDNA,大小约为75kb。Objective To explore the biological characteristics of a cold-active bacteriophage SWP-8against Flavobacteriaisolated from the Mingyong glacier area.Methods The morphology,restriction fragment length polymorphism(RFLP)of genomic DNA and physiological properties of the bacteriophage SWP-8were analyzed by using transmission electron microscopy(TEM)and CsCl density gradient centrifugation techniques.Results The bacteriophage SWP-8had an isometric head(about 150 nm in diameter)and a tail(about 150 nm in length and 20 nm in width),and a contractile sheath.The head and tail tube junction by a neck ring structure was detected.Physiological analysis showed that SWP-8had infection activity at 4 ℃;clear and transparent plaques were formed on double-layer plates between 4and 25 ℃.The optimal pH of infection was 7.0,and it had a wide range of pH tolerance.The optimum multiplicity of infection(MOI)was 10.000;the latency and lytic period were 60 min and 90 min,respectively,and it had high cracking efficiency.Conclusion SWP-8is a lytic Myoviridae bacteriophage.It is insensitive to chloroform and proteinase K.The genome of SWP-8is consisted of double-stranded DNA of approximately 75 kb.
