清热解毒化浊片对内毒素血症兔血浆内毒素、血常规及Toll样受体4/核因子-κB表达的影响

摘　　要：目的:探讨清热解毒化浊片对内毒素血症(ETM)模型兔血浆内毒素、血常规、肝脏组织TLR4(Toll样受体4)/NF(核因子)-κB的影响。方法:将21只健康新西兰大耳白兔随机分为正常组、模型组,清热解毒化浊片组(简称清化组)。模型组及清化组兔分别从耳缘静脉注入大肠杆菌内毒素10μg/kg造模,正常组兔注入10μg/kg生理盐水。造模2h后灌胃。结果:灌胃后2h,各组兔分别于心脏采血检测血浆内毒素、血常规,免疫组化法观察各组兔肝脏组织TLR4、NF-κB的表达。与模型组比,清化组免内毒素下降(P<0.05)而WBC(白细胞计数)上升(P<0.05),肝脏组织TLR4、NF-κB表达均下降(P<0.05)。结论:清热解毒化浊片对ETM兔血浆内毒素有明显清除作用,并可缓解大肠杆菌内毒素对WBC损害,可能下调TLR4/NF-κB信号通路。这或许是清热解毒化浊片治疗多种感染性疾病疗效的物质基础。Objective： To explore the effect of Qingre Jiedu Huazhuo tablet on expression of endotoxin, blood Routine TLR4 and NF-κB signaling pathway in ETM rabbits Methods：The 21 healthy New Zealand big-eared rabbits were randomly divided into normal group, model group and Qingre JieDu Hua zhuo Plan group （ Qing Hua group）. Model group and Qing Hua group were injected with10μg/kg Escherichia Coli Endotoxin from the rabbit ear vein respectively to set model and the normal group were injected with 10 g/kg saline. Then the rabbits were gavaged and plasma endotoxin, blood routine, expression of TLR4 and NF-kB were observed. Results： Compared with the model group, rabbit endotoxin , hepatic TLR4 and NF-kappa B in treatment group were decreased respectively （ P 〈 0.05 ） and WBC increased （ P 〈 0.05 ）. Conclusion：Qingre Jiedu Huazhuo tablet has obvious removal effect in ETM rabbit plasma endotoxin, and can help the WBC e. coli endotoxin damage ; It also can decrease the TLR4/NF-κB signaling pathway. This may be the material basis for the treatment of many kinds of infectious diseases.
