摘 要:应用MTT法检测黄连素联合Ta1722对SMMC-7721细胞活力的影响,以金正均法计算q值判断两药相互作用。根据细胞划痕实验与Hoechst染色实验,研究黄连素联合Ta1722对SMMC-7721的迁移及凋亡的影响。使用western blot检测迁移和凋亡相关蛋白表达。黄连素浓度在3.125~25μmol/L范围内、Ta1722浓度在20μmol/L时,两者为协同作用,黄连素联合Ta1722对SMMC-7721细胞的迁移无明显作用,迁移相关蛋白NF-κB,CXCR4,MMP2,MMP9表达无明显变化。黄连素联合Ta1722可诱导SMMC-7721细胞凋亡,上调P53,上调促凋亡蛋白Bax,下调抑凋亡蛋白Bcl-2。黄连素联合Ta1722对SMMC-7721细胞的迁移无明显作用,但可诱导SMMC-7721细胞凋亡,其机制可能是其上调抑癌基因P53的表达,从而导致促凋亡Bax上调,抑凋亡蛋白Bcl-2下调有关。Cell proliferation was assessed by MTT assay. The interaction of Berberine and Ta1722 was esti- mated by Jin Zheng jun's method. Wound Scratch assay and Hoechst assay were used to study the effect of Berber/ne combined with Ta1722 on apoptosis and migration of SMMC-7 721 liver cancer ceils. There is a synergic effect between the Berberine and Ta1722 when concentrations of Berberine change from 3. 125 μmol/L to 25 μmol/L and the concentration of Tal 722 is 20μmol/L. There is no significantly effect on cell migration, as well as NF-KB, CXCR4, MMP2 and MMP9 expression. However, Berberine combined with Ta1722 induced apoptosis which could upregulate P53 and Bax expression and downregulate Bcl-2 expression. Berberine combined with Ta1722 has no significantly effect on cell migration, but can induce apoptosis on SMMC-7721, this effect may be related to the increase of the expression of P53 which leads to the increase of the expression of Bax, reducing the expression of Bcl-2.
