摘 要:目的探讨Ro52表达水平是否与系统性红斑狼疮(systemic lupus erythematosus,SLE)患者B淋巴细胞的异常激活相关。方法分离14例SLE患者和10名健康对照的外周血单个核细胞,使用有丝分裂原美洲商陆(pokeweed mitogen,PWM),抗CD40,抗IgM+抗CD40,抗IgG+抗CD40刺激B细胞;碘化丙啶(propidium iodide,PI)染色流式细胞仪分析细胞周期,S期与G2/M期细胞为增殖细胞;定量PCR检测Ro52表达。结果 SLE患者B细胞激活后增殖水平显著高于健康对照(抗CD40、抗IgM+抗CD40、抗IgG+抗CD40刺激后,SLE患者与健康对照增殖细胞分别为3.6%±0.7%、7.9%±2.5%、5.1%±1.3%与2.4%±0.6%、3.7%±1.3%、2.7%±0.5%,P均<0.05),而Ro52表达水平显著低于对照(PWM、抗CD40、抗IgM+抗CD40、抗IgG+抗CD40刺激后,SLE患者与健康对照Ro52表达水平分别为0.50±0.09、0.92±0.16、0.84±0.19、1.03±0.21与1.41±0.20、3.05±0.64、2.19±0.35、3.10±0.72,P均<0.05)。Ro52表达水平与处于S和G2/M期的增殖细胞数量呈负相关,PWM、抗CD40、抗IgM+抗CD40以及抗IgG+抗CD40激活B细胞后Ro52表达水平与增殖细胞百分数的相关系数分别为-0.42、-0.25、-0.49和-0.40,P均<0.05。抗CD40激活的SLE患者B细胞比健康对照出现更加显著的Ro52表达水平下降。结论 Ro52表达水平在B细胞增殖时下调,可能参与SLE患者B细胞的异常激活。Objective To investigated whether the expression level of Ro52 was related to the abnormal activation of B cells in patients with SLE. Methods Peripheral blood mononuclear cells (PBMC) were isolated from 14 patients with SLE and 10 healthy controls. B cells were stimulated by pokeweed mitogen (PWM), anti-CD40 alone, anti-IgM plus anti-CD40 and anti-IgG plus anti-CD40. Cell cycle was analyzed by propidium iodide (PI) staining and flow cytometry, the cells in S stage or G2/M stage were proliferating cells. The expression of Ro52 was tested by quantitative real-time PCR using GAPDH as the internal control. Results After stimulated by anti-CD40, anti-IgM + anti-CD40, anti-IgG + anti-CD40, the percentage of proliferating B cells from patients with SLE was significantly higher than that of healthy controls, which was 3.6% ±0. 7%, 7.9%±2. 5%, 5.1% ± 1.3% versus 2.4% ±0. 6%, 3.7% ±1.3%, 2.7% ±0. 5%, respectively (P 〈 0.05 ) . However, the expression level of Ro52 by PWM, anti-CD40, anti-IgM + anti-CD40, anti-IgG + anti-CD40 stimulation was significantly declined, which was 0.50±0.09, 0.92±0.16, 0.84±0.19, 1.03±0.21 versus 1.41±0.20, 3.05 ±0.64, 2.19±0. 35, 3.10±0. 72 respectively (P 〈 0.05 ). The expression level of Ro52 was negatively correlated to the percentage of proliferating B ceils. After B ceils were activated by PWM, anti-CD40 alone, anti-IgM plus anti-CD40 or anti-IgG plus anti-CD40, the correlation coefficient of Ro52 expression and percentage of proliferating cell was - 0. 42, - 0. 25, - 0. 49 and - 0. 40, respectively, P 〈 0. 05. After being activated by anti-CD40, the Ro52 expression of B cells from patients with SLE showed significant decrease than that from healthy controls. Conclusion The expression of Ro52 is down-regulated when B cells are proliferating and this may be involved in the abnormal activation of B cells in patients with SLE.
