摘  要:目的:该研究以唾液斑为研究对象,研究碘熏、茚三酮和"502"熏显等三种痕迹显现方法对唾液斑生物检材DNA检测的影响,同时探讨该三种方法在唾液斑生物检材发现上的作用。方法:分别采用碘熏、茚三酮和"502"熏显法处理烧杯口唾液斑,采用棉签擦拭法采集烧杯口唾液斑DNA,采用chelex法提取DNA,Identifiler Kit试剂盒进行PCR扩增,3130基因分析仪进行扩增产物分型。结果与讨论:"502"熏显、碘熏对唾液斑DNA检测无明显影响,且"502"熏显在唾液斑生物检材发现上具有应用价值。茚三酮喷显对唾液斑DNA检测影响大,经茚三酮喷显处理的唾液斑难以进行有效DNA检测。Objective: the research aims to study the effect of three methods of trace showing, including Iodinesmoking, ninhydrin and “502”, on saliva spot DNA test, and discuss the application of the three methods in the detectionof saliva spot. Methods: the saliva spots leaving on the edge of beaker were deal with the three trace showing methodsrespectively, and then collected by cotton swabs. DNA was extracted by chelex method and PCR by Identifiler Kit. Finallythe PCR products were detected by 3130 Gene Analyzer. Results and Conclusions: “502” and Iodine somking have noeffect on the test of DNA on saliva spot and “502” is useful in the detection of saliva spot. Ninhydrin have significantlynegative effect on DNA test.
