摘 要:目的探讨红花黄色素(safflower yellow,SY)对离体大鼠心肌缺血/再灌注损伤(myocardium ischemia/reperfusion injury,MI/RI)的保护作用及其降低受体相互作用蛋白3(receptor interacting protein 3,RIB)硝化的可能机制。方法SD雄性大鼠36只,采用随机数字表法分为6组(每组6只):正常对照组(A组)、缺血,再灌注损伤(ischemia/reperfusioninjury,I/RI)组(B组)、SY干预组(C组,10μmol/L。;D组,30μmol/L;E组,90μmol/L)、阳性对照药依达拉奉组(F组,10μmol/L)。再灌注120min末,2,3,5.氯化三苯基四氮唑(2,3,5-triphenyl tetrazolium chloride,TYC)染色法测定心肌梗死面积百分比,免疫印迹法及免疫沉淀法测定RIB、受体相互作用蛋白1(receptor interacting protein1,RIP1)的表达水平及RIP3硝化水平、RIP1磷酸化水平,化学比色法测定心肌组织中超氧化物歧化酶(superoxide dismutase,SOD)活力、丙二醛(malonaldehyde,MDA)含量。结果与B组[(36.3±4.7)%]比较,D组[(15.3±4.2)%]、E组[(6.9±4.4)%]、F组[(9.6±1.2)%]心肌梗死面积减小(P〈O.05);与B组比较,D组、E组、F组SOD活性升高(P〈0.05),MDA含量降低(P〈O.05);各组RIP3及RIP1表达水平比较,差异无统计学意义(P〉0.05);与B组比较,D组、E组、F组RIP,硝化水平均降低(P〈O.05),RIP1磷酸化水平均降低(P〈0.05)。结论SY降低I/RI中的氧化水平,下调RIP3硝化及RIP1磷酸化水平,对MI/RI发挥保护作用。Objective To investigate the protective effects of safflower yellow (SY) pigment against ischemia/reperfusion injury (I/RI) of rat isolated hearts and the potential role of receptor interacting protein 3 (RIP3) nitration. Methods Thirty-six SD male rats were randomly divided into 6 groups (n=6): normal control group (group A), I/RI group (group B) , SY groups (group C, 10 μmol/L, group D, 30 μmol/L, group E, 90 μmol/L), edaravone group (group F, 10 μmol/L). At the end of reperfusion, the percentages of myocardial infarction area with 2, 3, 5-triphenyl tetrazolium chloride (TIC) staining method were measures after reperfusion. The expression of RIP3, receptor interacting protein I(RIPI), nitration of RIP3(N-RIP3) and phosphorylation of RIP1(P-RIPI) were measured by western blotting combined with immune precipitation. Activity of superoxide dismutase (SOD) and content of malonaldehyde (MDA) in myocardial tissue were measured with chemical colorimetry. Results At the end of reperfusion, compared with group S [(36.3±4.7)% ], area of myocardial infarction of group D[ (15.3±4.2)% ], group E [(6.9±4.4)% ] and group F[ (9.6±1.2)% ] decreased (P〈0.05). Compared with group B, SOD activity of myocardial tissue in group D, group E and group F were increased (P〈 0.05) and MDA content were lower (P〈0.05). At the end of the reperfusion, there were no differences in expression of RIP3 and RIP1 in myocardial tissues in each group (P〉0.05). Compared with group B, the level of N-RIP3 in group D, group E and group F decreased (P〈0.05), the level of P-RIP1 in group D, group E and group F also decreased(P〈0.05). Conclusions SY protects I/RI of rat isolated heart by down-regulating oxidation level and decreasing the level of N-RIP3 and P-RIP1 nitration.
