风湿宁胶囊含药血清干预对CIA-FLS细胞增殖及凋亡的影响

摘　　要：目的:探讨风湿宁胶囊(FSN)含药血清干预对CIA-FLS细胞增殖及凋亡的影响。方法:采用CCK-8法检测各组血清干预24、48、72 h后对CIA-FLS细胞增殖状态的影响;采用Annexin V-FITC/PI双染法通过流式细胞仪观察各组血清干预48 h后对CIA-FLS细胞早期和晚期凋亡情况的影响。结果:CCK-8法结果表明,在24 h,FSN各浓度(20%、10%和5%)含药血清干预对CIA-FLS细胞的增殖无抑制作用;在48 h,20%FSN含药血清和10%FSN含药血清干预对CIA-FLS细胞的增殖都表现出不同程度的抑制。在72 h,FSN各浓度(20%、10%和5%)含药血清对CIA-FLS细胞增殖均可见显著的抑制效果,其中20%风湿宁胶囊含药血清效果最好。Annexin V-FITC/PI双染检测结果表明,风湿宁胶囊含药血清各浓度(20%、10%和5%)组都不同程度地提高了细胞总凋亡率,其中20%风湿宁胶囊含药血清组对CIA-FLS细胞凋亡促进作用最强。结论:风湿宁胶囊含药血清对CIA-FLS细胞增殖的抑制及凋亡的促进可能是其治疗类风湿关节炎的机制之一。Objective： To research the effects of FSN drug-containing capsule serum on CIA-FLS cell proliferation and apoptosis. Methods： CCK-8 method is used to detect serum intervention group 24,48,72 h after the impact on the CIA-FLS cell proliferation state; The Annexin V-FITC / PI double staining with flow cytometry instrument observation group serum intervention after 48 h of CIA-FLS the impact of early and late apoptotic cells. Results： The CCK-8 method detection results show that within 24 h,the concentration（ 20%,10% and 20%） of FSN drug-containing serum on the CIA-FLS did not appear obvious role. In 72 h,FSN each concentration（ 20%,10% and 5%） drug-containing serum and tofacitinib drugcontaining serum on the CIA-FLS proliferation had visible significant inhibitory effect,of which 20% Fengshining capsules containing medicine serum works best. Annexin V-FITC / PI double dye test results show that the Fengshining capsules containing medicine serum concentrations（ 20%,10% and5%） groups each have different levels of the cell apoptosis rate,of which 20% Fengshining capsules containing medicine serum group of CIA-promote the strongest FLS apoptosis.Conclusion： Fengshining capsule drug-containing serum on the CIA-FLS inhibition of cell proliferation and apoptosis of promotion may be one of the mechanism of treatment of rheumatoid arthritis.
