摘 要:目的:探讨芪蟾口服结肠靶向片中有效组分(ECOQCOT)联合5-氟尿嘧啶(5-FU)干预治疗对小鼠CT26结肠癌移植瘤的作用,及对小鼠外周血中T淋巴细胞亚群和移植瘤组织中凋亡相关蛋白表达的影响。方法:建立CT26结肠癌细胞小鼠皮下移植瘤模型,随机分为模型组,提前干预组,同步干预组,延后干预组,5-FU治疗(20 mg·kg^(-1))组,ECOQCOT治疗(22g·kg^(-1))组。绘制各组小鼠移植瘤生长曲线,计算抑瘤率;流式细胞术检测外周血中T淋巴细胞亚群表达;免疫组化法检测各组移植瘤组织中B细胞淋巴瘤-2(Bcl-2)和Bcl-2相关X蛋白(Bax)蛋白表达。结果:提前干预组,同步干预组,延后干预组,5-FU治疗组,ECOQCOT治疗组抑瘤率分别为43.63%,34.35%,31.12%,26.91%,18.67%。与模型组比较,各用药组CD3^+,CD4^+,CD8^+细胞比例及CD4^+/CD8^+均升高,其中提前干预组和同步干预组CD3^+,CD4^+,CD4^+/CD8^+均显著高于模型组(P<0.01)。免疫组化结果显示,与模型组比较,提前干预组、同步干预组Bax蛋白表达明显升高(P<0.05,P<0.01);提前干预组,同步干预组,延后干预组,5-FU组Bcl-2蛋白表达均明显降低(P<0.05,P<0.01)。结论:各ECOQCOT联合5-FU组抑瘤作用明显强于单一5-FU组和单一ECOQCOT组,并以提前干预组抑瘤作用最强;其机制可能与增强机体的免疫功能和诱导肿瘤细胞凋亡有关。Objective: To explore the effects of effective components of Qi-toads colon-specific oral tablets( ECOQCOT) combined with 5-fluorouracil( 5-FU) on CT26 colon carcinoma transplanted tumors,and investigate the T-lymphocyte levels in the peripheral blood and the protein expression of related apoptosis regulatory factors in tumour tissues. Method: Subcutaneous transplanted tumor models were established in CT26 tumourbearing mice,and then were randomly divided into model group,early intervention group,synchronous intervention group,delayed intervention group,5-FU group( 20 mg·kg^(-1)),and ECOQCOT group( 22 g·kg^(-1)). The growth curves of transplanted tumors were drawn and tumour inhibition rate was calculated in each group. the Tlymphocyte levels in the peripheral blood were detected by flow cytometry,the protein expressions of Bax and Bcl-2were detected by immunohistochemical staining. Result: The tumour inhibition rate was 43. 63%,34. 35%,31. 12%,26. 91% and 18. 67% respectively in early intervention group,synchronous intervention group,delayed intervention group,5-FU group and ECOQCOT group. As compared with the model group,the percentage of CD3^+,CD4^+,CD8^+T cells and CD4^+/CD8^+were increased in all treatment groups,and the above indexes in early intervention group and synchronous intervention group were significantly increased( P〈0. 01). The results of immunohistochemical staining showed that the protein expressions of Bax in early intervention group and synchronous intervention group were significantly higher than those in model group( P〈0. 05,P〈0. 01),the protein expressions of Bcl-2 in early intervention group,synchronous intervention group,delayed intervention group and 5-FU group were significantly lower than those in model group( P〈0. 05,P〈0. 01). Conclusion:ECOQCOT combined with 5-FU had stronger inhibiting effects on CT26 tumours than the single 5-FU group or single ECOQCOT group,and the early intervention group had the strongest inhibit
