摘 要:目的:探讨降糖消渴颗粒对高脂饮食诱导2型糖尿病KKAy小鼠糖脂代谢的影响。方法:30只KKAy小鼠予以高脂饲料喂养4周后,建立2型糖尿病(空腹血糖≥13.9 mmol·L^(-1))小鼠模型。将成模后的KKAy小鼠随机分为模型组、吡格列酮组、降糖消渴颗颗粒高、中、低剂量组(按生药量计为1.75、3.5、7 g·kg^(-1)),6只C57BL/6J鼠为正常对照组。干预10周后,称取体质量和肝重,计算肝重指数;检测小鼠血清和肝脏中甘油三酯、胆固醇、高密度脂蛋白、低密度脂蛋白的含量。结果:降糖消渴颗粒低剂量(1.75 g·kg^(-1))可显著降低血清总胆固醇(TC)、低密度脂蛋白胆固醇(LDL)(P<0.01),且可降低肝脏中甘油三酯(TG)含量(P<0.05);中剂量(3.5g·kg^(-1))显著降低了糖尿病小鼠的肝重指数,对血清和肝脏脂质各项指标都具有良好的改善作用(P<0.01);高剂量(7 g·kg^(-1))不仅降低了糖尿病小鼠的肝重指数(P<0.01),对血清中TG、LDL及肝脏脂质各项指标也有显著改善。结论:降糖消渴颗粒可以有效改善T2DM状态下肝脏的脂质代谢紊乱,降低血脂含量。Objective:To investigate the effect of Jiangtang Xiaoke Granule (JTXKG) on serum and hepatic lipids me-tabolism in type 2 diabetic KKAy mice which were induced by high fat diet. Method: A total of 30 KKAy mice were fed by high - fat 5 diet for 4 weeks. T2DM model was established when the FBG ≥ 11.1 mmol .L - 1. The T2DM mice were randomly divided into model group,pioglitazone group and JTXK granule groups in low (1.75 g/kg), medium (3.5 g/kg) and high (7 g/kg)doses. The normal group had 6 C57BL/6J mice in it. After 10 -weektreatment,the body weight,he- patic weight, hepatic index, serum and hepatic TG, TC, HDL and LDL were measured. Result : Compared with model group, T2DM mice supplemented with 1.75 g/kg dose of JTXK granule lowered serum TC and LDL ( P 〈0. 01 ) and hepatic TG (P 〈 0. 05). T2DM; mice fed with 3.5 g/kg dose of JTXK Granule induced hepatic index (P 〈 0. 01 ) and lipid in both serum and liver (P 〈0: 01). T2DM mice administrated with 7 g/kg dose of JTXK Granule lowered hepatic index ( P 〈 0. 01 ) and serum TG, LDL and hepatic lipid were significant. Conclusion:JTXKG improved hepatic lipid metabolism in T2DM mice and lowered lipid in serum.
