补肾中药对PTSD孕鼠子代行为遗传及血清皮质酮的干预研究

摘　　要：目的：观察PTSD孕鼠子代行为遗传及补肾中药金匮肾气丸对“胎损”子代行为及血清皮质酮的干预效果。方法：将初次受孕成功的36只SD大鼠随机分为空白组、PTSD组、中药组，每组12只。各组孕鼠于妊娠第7—13天采用单程长时应激法进行创伤应激造模，空白组除外。造模后，中药组孕鼠给予添加金匮肾气丸的补肾中药饲料至妊娠结束。观测各组子代鼠出生当天及第5、15、25天体质量、行为能力及外周血皮质酮水平。结果：PTSD组出生体质量及体质量增长、翻身能力、旷场试验得分低于中药组和空白组（P〈0．05），血清皮质酮浓度高于其余两组（P〈0．05）；除体质量增长外，中药组和空白组之间无差异（P〉0．05）。结论：孕鼠PTSD恐惧应激可遗传到子代，致子代低出生质量、体质量增长缓慢，行为能力及血清皮质酮异常。补肾中药金匮肾气丸可纠正上述异常改变，减少对子代生长发育及神经内分泌损害。Objective： To observe behavioral genetics of PTSD and explore the effect of Jin Kui Shen Qi pill on behavior change and serum corticosterone level of offspring from pregnant rats with post - traumatic stress disorder（PTSD）. Meth- ods： 36 pregnant rats were randomized to three groups： control group （CG）, PTSD group （PG） and herbal group （HG）. There were 12 rats in each group. At the second trimester, PTSD rat models were made by Single -prolonged stress （SPS） except in CG, which has been extensively applied in PTSD research. Jin Kui Shen Qi pill was given to HG in their feeds after the models were established. The body weight （0,5,15,25 d）, behavior test of survived offspring were measured and observed. Serum corticosterone was detected by ELISA method. Results： The average birth weight, weight gain, ability to roll over and open - field test＇ performance of offspring in PG were significantly lower than in HG and CG （P 〈 0. 05 ）. The level of serum cortieosterone was higher than the other two groups（ P 〈 0. 05 ）. No statistics differences was observed between CG and HG except in weight gain （ P 〉 0.05 ）. Conclusions ： The negative influence derived from maternal P＇PSD could be handed down to the offspring resulting in low birth weight, growth retardation, also leading to ab- normal behavior and serum corticosterone disturbance. Jin Kui Shen Qi Pill could improve above - mentioned so as to pro- tect the baby rats from neuroendocrine damage caused bv maternal stress condition.
