摘 要:目的:探讨益气补肾方法对于乳腺癌骨转移的抑制作用及其作用的机制。方法:将雌性Balb/c裸鼠随机分为治疗组和对照组,采用人乳腺癌细胞株MDA-MB-231注射裸鼠左心室,制作乳腺癌动物模型,分别予以益气补肾中药及生理盐水口服,通过骨ECT检查及病理检查明确其骨转移情况,然后通过免疫组化法,分别测定每组裸鼠血清中BSP、OPN,MMP-1的表达情况。结果:对照组的裸鼠骨转移发生率为80%,而治疗组的发生率为46.67%,与对照组比较,治疗组中裸鼠骨转移的发生率要明显降低;通过免疫组化检测,治疗组中的MMP-1和OPN的表达要明显低于对照组(P<0.05),治疗组中BSP的表达也要明显低于对照组(P<0.01)。结论:益气补肾中药能有效减少乳腺癌骨转移裸鼠模型的骨转移发生率,其作用机制可能为:通过对MMP-1,BSP,OPN的表达抑制,减少了乳腺癌细胞的脱落、迁移,并最终通过OPG-RANKL-RANK系统,抑制了破骨细胞的分化和骨的重吸收,减少了乳腺癌的转移和骨组织的破坏。Objective : To observe the effects and study the mechanism of Yiqibushen on breast cancer bone metastasis in mice. Methods: The female Balb/c nude mice were randomly divided into treatment and control groups, and then injec- ted with MDA -MB -231 cells into the left ventricles. The treatment and control groups were fed with Yiqibushen Chi- nese medicine and normal saline separately, and then we observed the bone metastasis after the bone ECT scanning and pathological examination. Finally, the expressions of the BSP, OPN and MMP - 1 were detected in each group by using the IHCA. Results : The control groups' rate of bone metastasis was 80% while the treatment group's rate was 46.67%. The rate of the treatment group was obviously lower than the control group' s. The expressions of MMP - 1 and OPN in treat- ment groups were obviously lower than those in the control groups (P 〈 0.05) and the expression of BSP in the treatment group was obviously lower too ( P 〈 0.01 ). Conclusions : Yiqibushen medicine can reduce the bone metastasis in breast cancer mice effectively. Its possible mechanism was to reduce the shedding and migration of the breast cancer cells by down - regulating the expressions of MMP - 1, BSP and OPN and inhibiting the osteoelast differentiation and bone reab- sorption and reducing the bone metastasis and bone destruction through the OPG - RANKL - RANK system.
