加味四逆散对大鼠酒精性脂肪肝的实验研究

摘　　要：目的:观察加味四逆散对酒精性脂肪肝大鼠血清ALT、AST及肝组织CYP2E1及TNF-α的影响。方法:将72只大鼠随机分为3组,对照组、模型组、治疗组各24只。模型组和治疗组用乙醇灌胃的方法造模,而对照组应则采用蒸馏水。治疗组在4周时灌胃酒2 h前灌胃加味四逆散1 m L/100 g,模型组灌胃等量蒸馏水。共计12周。观察各组ALT、AST及肝组织SOD、MDA及CYP2E1和TNF-α的表达。结果:与模型组比较,治疗组血清ALT、AST,肝组织CYP2E1和TNF-α的表达下降,差异有统计学意义(P<0.05),MDA水平减少(P<0.05),SOD水平升高(P<0.05)。结论:加味四逆散对大鼠酒精性脂肪肝有一定疗效,通过抑制CYP2E1和TNF-α的表达,减轻脂质过氧化反应,可能是其作用的靶点之一。Objective ： To observe the effect of Jiawei Sini Powder on serum levels of ALT and AST, the expressions of cytochrome P4502E1 and tumor necrosis factor - αin the liver tissue of alcoholic liver disease（ALD） rats. Methods ： Totally 72 rats were randomly divided into the control group（ n = 24）, the model group （n = 24） and the Jiawei Sini Powder group （n = 24）. The rats model was prepared by given alcohol by gavage while distilled water was intragastrically given to those in the control group. Extract of Jiawei Sini Powder was intragastrically given to rats in the rest two groups. The ther- apeutic course lasted for 12 weeks. Serum levels of ALT and AST, liver pathology and the expressions of SOD, MDA, CYP2E1 and TNF - α in the liver tissue of ALD rats were observed after administration of Jiawei Sini Powder. Results ： Compared with the model group, serum levels of ALT and AST decreased. The liver pathology was improved and the expressions of CYP2E1 and TNF - α in the liver tissue decreased, showing statistical difference （P 〈 0.05 ）. The levels ofMDA in liver tissue of model group were decreased significantly （ P〈 0.05 ） and the level of SOD was increased signifi- cantly. Conclusion： The results demonstrate that Jiawei Sini Powder has certain effect on treating ALD and reducing lipid peroxidation. The mechanisms of this effect may involve its inhibition on CYP2E1 and TNF- α.
