摘 要:目的:观察清上蠲痛合剂对偏头痛大鼠的预防作用及对血清中β-内啡肽(EP)、降钙素相关基因肽(CGRP)、一氧化氮(NO)的影响。方法:分组方法及给药方式采用硝酸甘油(GTN)皮下注射法建立偏头痛大鼠模型,将成年健康SD大鼠36只,雌雄各半,体质量(188.29±7.60)g,随机分为正常组(生理盐水剂量:与等效剂量等容积),模型组(生理盐水剂量:与等效剂量等容积),清上蠲痛合剂预防组[等效剂量,7.46 g/(kg·d)],以上每组12只。预防组及模型组连续给药6 d后造模用药。除正常组外,其余各组采用皮下注射硝酸甘油方法复制大鼠偏头痛模型,观察其耳红、甩头或挠头、爬笼出现时间、次数及持续时间等行为学指标后,采用酶联免疫法吸附法(ELISA)测定各组大鼠血清中EP、CGRP、NO的含量。结果:与正常组相比,造模后模型组大鼠血清β-EP含量较正常组下降(P<0.001);清上蠲痛合剂预防组大鼠血清中β-EP含量较模型组明显升高(P<0.05)。造模后模型组大鼠血清CGRP和NO含量较正常组升高(P<0.001),清上蠲痛合剂预防组能抑制上述改变(P<0.05)。结论:清上蠲痛合剂能明显预防大鼠偏头痛发作,可能与调节机体CGRP、NO、EP的释放有关。Objective:To observe the preventive effect of Qingshang Juantong Decoction on rats with migraine and the effect on β - endorphin ( EP ), calcitonin gene related peptide ( CGRP ) and nitric oxide ( NO ) in serum. Methods : Grouping method and administration: glyceryl trinitrate (GTN) subcutaneous injection for the establishment of rat model of migraine. A total of 36 adult healthy SD rats, half male and half female, weight (188.29g ± 7.60)g,were randomly as- signed to normal group ( saline dose with the equivalent dose volume) , model group ( saline dose with the equivalent dose volume) and Qingshang Juantong Decoction prevention group [ equivalent dose,7.46g/( Kg · d) ]. There were 12 rats in each group. The prevention group and model group were continuously administered for 6 days after modeling medication. Except the normal group, the rest groups by subcutaneous injection of nitroglycerin method duplicated the rat models of migraine. Observe the red eared, shakes or scratching their heads, climbing cage appeared time, frequency and duration of behavior index. By enzyme linked immunosorbent assay adsorption method (ELISA) ,we determined the contents of β- EP, CGRP and NO in serum. Results : Compared with the normal group, after modeling, model group rats serum β - EP decreased significantly compared with that of the normal group (P 〈 0. 001 ). The Qingshang Juantong Decoction prevention group' s β- EP was significantly increased compared with that of the model group ( P 〈 0. 05 ). After comparing with the normal group, the model group' s CGRP and NO were increased significantly (P 〈 0. 001 ) and Qingshang Juantong Decoction prevention group can inhibit the above changes ( P 〈 0. 05 ). Conclusion : Qingshang Juantong Decoction can obviously prevent migraine,which may be related to regulating the release of CGRP, NO and EP.
