水稻线粒体特异序列在细胞质多态性鉴定中的应用

摘  要:为了开发一套用于水稻细胞质多态性鉴定的分子标记,本研究通过对已测序的4个水稻线粒体基因组CW-CMS、LD-CMS、Nipponbare和9311线粒体基因组进行比较分析,获得55个大于100 bp的线粒体特异序列,并开发出一套水稻线粒体特异序列分子标记。该套分子标记稳定可靠,在对随机选择的23个水稻品种的线粒体组型鉴定中,共有24个线粒体特异标记扩增出多态性,使用UPGMA法聚类分析,在相似系数为0.46,可以将23水稻品种分为2大类群,其中第一大群可分为两个小类群,类群Ⅰ为粳稻聚类群,该类群大部分为粳稻品种,同时也有2个籼稻品种,推测这两个品种细胞质来源于粳稻;类群Ⅱ包含两个籼稻品种,与粳稻的细胞质亲缘关系较近;而第二大聚类群全部为籼稻品种,大部分品种间多态性丰富。因此,该套水稻线粒体特异序列标记可用于水稻细胞质多态性鉴定分析,同时该类型分子标记的开发对于其他物种的细胞质多态性鉴定具有指导意义。In order to develop a set of molecular markers for the identification of cytoplasmic polymorphism, four sequenced mitochondrial genomes of rice CW-CMS, LD-CMS, Nipponbare and 9311 were compared, 55 specific sequence above 100 bp were detected, which were translated into a set of rice mitochondrial specific markers. The markers were stable and reliable, 23 rice varieties randomly selected were PCR amplification analyzed, 24 markers showed polymorphism, based on which, the rice varieties could be divided into two categories groups (similarity coefficient 0.46) using UPGMA, one of which contained two subgroups. Subgroup I were Japonica group, most of which were Japonica, only two Indica varieties were speculated originated from Japonica. The cytoplasmic of the two Indica varieties in subgroup II were close to Japonica. All of the other group were Indicas, Polymorphism were rich among the most varieties. So, the set of markers could be used to identify rice cytoplasmic polymorphism, while the way to develop mitochondrial specific markers could be emulated for cytoplasmic identification in other species.
