摘 要:目的分析矮地茶多糖中的单糖组成及摩尔比值。方法采用硫酸水解多糖,1-苯基-3-甲基-5-吡唑啉酮柱前衍生化高效液相色谱法和薄层色谱法同时分析矮地茶多糖的单糖组成和摩尔比,Agilent TC-C18色谱柱,流动相:乙腈-p H为7.0的磷酸缓冲液(20∶80,0.1%),检测波长:250 nm,流速:1 m L/min;柱温:25℃。结果薄层色谱法和高效液相色谱法结果皆表明矮地茶多糖由葡萄糖(Glu)和半乳糖(Gal)组成,高效液相色谱法显示矮地茶多糖中Glu和Gal的摩尔比值约为1∶2.36。结论薄层色谱法可用于矮地茶多糖的单糖组成的快速鉴别,方法快速、准确;高效液相色谱法可用于矮地茶多糖中单糖组成的鉴别和摩尔比值测量,方法简单、灵敏、分离效率高。Objective To analyze the monosaccharide composition and the molar ratio of Ardisia japonica Polysaccharide (AJP), Methods Sulfuric acid was used to hydrolyze AJP. Pre-column 1-Phenyl-3-methyl'5-pyrazalone (PMP) derivation HPLC and TLC method were developed to determine the monosaccharide compositions and the molar ratio of AJP. The mobile phase was acetonitrile-pH 7.0 phosphate buffer (20:80, 0.1%) with Agilent TC-C18 column. The detection wavelength was 250 nm, the flow rate was 1 mL/min, and the column temperature was 25 ℃. Results The resuits of TLC and HPLC analysis showed that AJP were consisted of glucose (Glu) and galactose (Gal). Meanwhile HPLC analysis showed that the molar ratio of AJP was 1:2.36. Conclusion TLC which is fast and accurate, can be used for the rapid identification the monosaccharide composition in AJP. HPLC which is simple, sensitive and high separation efficiency, can be used for the quantitative analysis of the monosaccharide composition in AJP.
