摘  要:目的研究大孔树脂分离纯化塞北紫堇总生物碱的工艺条件.方法以盐酸小檗碱为对照品,采用酸性染料比色法,分别考察总生物碱在5种类型的大孔树脂(AB-8、NKA-9、LKY131、HP20、PHD400A)上的吸附和解吸附行为,同时分析了上样液浓度、洗脱液浓度和洗脱量、洗脱流速等参数对分离的影响.结果所比较的5种树脂中,AB-8型大孔树脂对总生物碱分离纯化效果最好,富集能力强,较优富集工艺为:2.0mg/ml上样液浓度、9倍量70%浓度的乙醇溶液洗脱、洗脱流速为2.0ml/min,最大吸附量为160mg/g.结论使用AB-8型大孔树脂对塞北紫堇总生物碱具有较好的纯化作用,可使其纯度提高16.6%.purpose To study the purification technology of total alkaloids from Corydalis impatiens(Pall.)Fisch by macroporous resin.Method Berberine hydrochloride as the control sample,which used acid dye colorimetry.The adsorption and desorption behavior of the total alkaloids in 5types of macroporous resins(AB-8,NKA-9,LKY131,HP20,PHD400A)were investigated.And the sample concentration,elution concentration,elution volume,elution velocity and other parameters,which were analyzed on the separation effect.Result The comparison of the 5kinds of resins,AB-8macroporous resin had the best effect and high enrichment ability for separation and purification of total alkaloids fromCorydalis impatiens(Pall.)Fisch.The optimum conditions were as follows:with 2.0mg/ml as eluant sample concentration,70% ethanol and 9times the amount of column volume as eluant,eluting at a flow rate2.0ml/min,maximum adsorption capacity of 160mg/g.Conclusion AB-8macroporous resin could purify total alkaloids fromCorydalis impatiens(Pall.)Fisch and improved the purity of 16.6%.
