摘 要:目的:建立测定雷公藤药材中雷公藤甲素含量测定方法,比较不同采收年份雷公藤药材中雷公藤甲素的含量差异。方法:用高效液相色谱法,以十八烷基键合硅胶为填充剂,以0.05 mol/L的磷酸二氢钠溶液-乙睛-甲醇(体积比为60∶15∶25)为流动相,采用梯度洗脱,检测波长218 nm,测定雷公藤甲素的含量。结果:高效液相色谱法测定雷公藤甲素进样量在0.044 25μg^0.59μg范围内线性良好,r^2=0.999 9,平均加样回收率为99.44%,RSD值为1.7%。结论:该方法准确、可靠、简便、重复性好,可用于雷公藤药材的质量控制。Objective: To establish the method for the content determination of triptolide in tripterygium wilfordii,and to compare the difference of the content of the triptolide in Tripterygium wilfordii collected in different harvest years. Methods:HPLC was adopted with octadecyl bonded silica as the filler,0.05mol/L of sodium dihydrogen phosphate-acetonitrile-methanol with the volume ratio of 60:15:25 as the mobile phase,and using gradient elution and detection wavelength of218 nm to determine the content of triptolide. Results:In the range of 0.044 25 μg ~0.59 μg,the linear relationship of triptolide was food,r^2=0. 999 9,the average spotting recovery rate was 99.44%,and RSD=1.7%. Conclusion:This method is accurate,reliable,simple and repeatable,and it can be used in the quality control of tripterygium wilfordii.
