摘 要:[目的]研究肛周注入白芍注射液对于大鼠肛直肠静息压力和收缩压力的影响以及可能的作用机制。[方法]48只Wistar大鼠随机分为3组,分别在肛周肌肉注入等量的白芍注射液、阿托品注射液和生理盐水,在注药前及注药后6h和72h分别测量肛直肠的静息压力和最大收缩压力,处死大鼠,在40倍光学显微镜下观察实验动物的肛门括约肌肌纤维组织形态学特点,并用ELISA法测量肌动蛋白的浓度,3组间以及每组前后进行比较。[结果]1静息压力:组内比较:白芍组注药后6h、72h相似,但均比注药前明显降低;阿托品组注药后6h比注药前和注药后72h均低,但注药后72h与注药前比较差异无统计学意义;生理盐水组无明显变化。组间比较:注药后6h,白芍组与阿托品组之间比较差异无统计学意义,但均比生理盐水组低;注药后72h,白芍组比其他2组均低,但阿托品组与生理盐水组比较差异无统计学意义。2收缩压力:组内比较:白芍组3个时间点比较差异无统计学意义;阿托品组:注药后6h比注药前和注药后72h均低,但注药后72h与注药前比较差异无统计学意义;生理盐水组3个时间点比较差异无统计学意义。组间比较:注药后6h,白芍组与生理盐水比较差异无统计学意义,但都比阿托品组高;注药后72h,3组间比较差异无统计学意义。3从组织学观察来看,白芍注射液和阿托品均能使肛门括约肌肌纤维结构较生理盐水组松散。4ELISA检测结果:白芍组与阿托品组比较,以及阿托品组与生理盐水组比较,差异均无统计学意义;白芍组与生理盐水组比较,差异有统计学意义。[结论]白芍注射液可以降低大鼠肛直肠的静息压力,而且比阿托品更加持久,但不能降低其最大收缩压力,其机理可能与肌动蛋白有关。[Objective]To observe the effect of Paeonia lactiflora Pall injection on anal sphincter rest and squeeze pressures of mouse,and to research the mechanism about it.[Methods]48mice were divided into 3groups and respectively injected into anal sphincter with Paeonia lactiflora Pall injection(PLPI),atropine(ATR),and NS.Measure the rest pressure(RP)and squeeze pressures(SP)at three time nodes(before injection,6hours later or 72 hours later after injection).The histology changes and actin expression in anal sphincter tissue were observed.[Results]1.RP:①RP of 6hours after and 72 hours in PLPI are similar,but both lower than before injection;RP of before injection and 72 hours after in ATR are similar,but both Higher than 6hours later;There were no differences in NS group among three time nodes.②At the time node of 6hours later,there was no difference between PLPI group and ATR group,but they were both lower than in NS group.At the time node of 72 hours later,there was no difference between NS group and ATR group,but they were both higher than in PLPI group.2.SP:①there were no differences in SP in PLPI group among three time nodes;SP of before injection and 72 hours after in ATR are similar,but bothHigher than 6hours later.There were no differences in NS group among three time nodes.②At the time node of 6hours later,there was no difference between NS group and PLPI group,but they were both Higher than in ATR group;At the time node of 72 hours later,there were no differences among three groups.[Conclusion]PLPI can weaken anal sphincter RP of mouse,but can't weaken SR.It's perhaps associated with actin in anal sphincter muscle.
