摘  要:目的探讨脊髓神经元自噬与大鼠糖尿病神经病理性疼痛(DNP)的关系。方法选取成年雄性健康SPF级SD大鼠60只,随机分为造模组50只和对照组10只。比较两组不同时间点血糖、机械疼痛阈值、LC3/TUNEL阳性神经元细胞数、LC3-Ⅱ蛋白表达和LC3-Ⅱ/LC3-Ⅰ比值的差异,以及各指标间的相关性。结果造模后,造模组血糖值随时间而升高,且高于对照组(P<0.05),机械阈值及热刺疼痛阈值均随时间而降低,且低于对照组(P<0.05);LC3/TUNEL阳性神经元细胞数,LC3-Ⅱ蛋白表达和LC3-Ⅱ/LC3-Ⅰ比值随时间而升高,且在不同时间点均高于对照组(P<0.05)。相关分析显示,血糖与LC3/TUNEL阳性神经元细胞数、LC3-Ⅱ蛋白表达、LC3-Ⅱ/LC3-Ⅰ比值呈正相关(P<0.05),机械疼痛阈值与LC3/TUNEL阳性神经元细胞数、LC3-Ⅱ蛋白表达、LC3-Ⅱ/LC3-Ⅰ比值呈负相关(P<0.05)。结论大鼠DNP模型中脊髓神经元自噬功能增强,提示其可能与DNH有关。Objective To investigate the relationship between spinal cord neuron autophagy and diabetic neuropathic pain in rats. Methods A total of 60 adult male healthy SD grade SPF rats were randomly divided into two groups., model group (n = 50) and control group (n = 10). Blood glucose, mechanical pain threshold, LC3/TUNEL positive neuron cell numbers, expression of LC3-Ⅱ and LC3-Ⅱ/ LC3-Ⅰ ratio at different time points were compared between the two groups. And correlation between these indexes was also analyzed. Results The blood glucose level increased with time in model group and was higher than in control group (P〈0. 05). The mechanical and spurs pain threshold decreased with time in model group and was lower than in control group (P〈0.05). The number of LC3/TUNEL positive neuron cells, expression of LC3- Ⅱ and LC3-Ⅱ/LC3- Ⅰ ratio at different time points increased with time in model group and were significantly higher in model group than in control group (P〈0.05). Correlation analysis showed that the blood glucose was positively correlated with the number of LC3/ TUNEL positive neurons, the expression LC3-Ⅱ, and LC3- Ⅱ/LC3- Ⅰ ratio ( P 〈 0. 05 ) . And the mechanical pain threshold was negatively correlated with the number of LC3/TUNEL positive neurons, the expression LC3- Ⅱ, and LC3-Ⅱ/LC3- Ⅰratio(P〈0.05). Conclusion The autophagy function of spinal cord neurons are enhanced in rat DNP model, suggesting that it may be related to the diabetic neuropathic pain.
