摘 要:目的观察电针对高脂喂养大鼠主动脉内皮细胞c-Jun氨基末端激酶(JNK)蛋白的影响。方法将24只雄性SD大鼠随机分为空白组、模型组、电针组8只。空白组予普通饲料喂养,模型组与电针组予高脂饲料喂养。造模8 w后,空白组、模型组继续如前喂养不进行干预,电针组予针刺双侧内关、三阴交、足三里及肾俞,双侧足三里及三阴交加电。干预4 w后,用葡萄糖氧化酶法、ELISA及Western印迹检测并比较各组大鼠空腹胰岛素(FINS)、空腹血糖(FPG)、稳态模型评价的胰岛素抵抗(IR)指数(HOMA-IR)以及主动脉内皮细胞JNK蛋白的表达及其磷酸化水平。结果与模型组比较,电针组FPG、FINS、HOMA-IR均显著降低(P<0.01),而与空白组比较差异不显著(P>0.01);与模型组相比较,电针组的JNK蛋白表达及其磷酸化水平显著降低(P<0.01),与空白组比较差异不显著(P>0.01)。结论电针治疗能调节血管内皮细胞JNK蛋白表达及其磷酸化水平,使其降至正常水平,从而改善IR大鼠的IR状态,增强IR大鼠胰岛素的敏感性。Objective To investigate the effect of electroacupuncture (EA) on the expression of JNK protein in aorta of rats with high fat induced insulin resistance. Methods 24 male SD rats were randomly divided into blank, model and EA groups, with 8 rats in each group. Rats in blank group were fed with normal diet, rats in model and EA groups were fed with high fat diet. After 8 weeks, rats in EA group were applied to bilateral Neiguan ( PC6 ), Zusanli ( ST36 ), Sanyinjiao ( SP6 ) and Shenshu ( BL23 ) for 4 weeks. Needles in Zusanli ( ST36 ), Sanyinjiao (SP6) were connected with EA. At the end of the treatment, glucose oxidase method and ELISA were used to detect the fasting plasma glucose (FPG) , fasting serum insulin(FINS) and homeostasis model assessment-insulin resistance (HOMA-IR)was calculated. The expression of JNK protein and the phosphorylation protein were determined by using Western blot. Results Compared with model group, the FPG, FINS and HOMA-IR in the EA group were significantly decreased( P〈O. 01 ). After treatment, the FPG, FINS and HOMA-IR in the EA group were sig- nificantly lower than those of model group( P〈0.01 )and the expression of p-JNK/JNK of rats in EA group was significantly decreased than that of model group(P〈0. 01 ). Condusions EA could improve insulin sensitivity of rats with high fat induced insulin resistance, which might be related to regulating the expression of JNK protein and its phosphorylation level.
