摘 要:目的:建立鸦胆子油的HPLC-ELSD指纹图谱分析方法,并与掺伪鸦胆子油指纹图谱进行比较。方法:运用SSI高效液相色谱仪和奥泰2000蒸发光检测器,采用Waters C_(18)(200 mm×4.6 mm,5μm)色谱柱,流动相:乙腈-二氯甲烷(60∶40),流速:1.2 ml·min^(-1),检测温度:85℃,柱温:40℃,进样量:15μl。结果:方法的精密度、重复性与稳定性良好,相对保留时间和相对峰面积的RSD均<3.0%,各峰分离度良好,指纹信息完整,符合指纹图谱技术规范。在此方法的基础上,建立了鸦胆子油及掺伪鸦胆子油HPLC-ELSD指纹图谱,并将相似度进行比较。结论:本试验为鸦胆子油质量控制和市场监管提供了重要依据。Objective: To establish an HPLC-ELSD fingerprint analysis method for Brucea javanica oil, and compare with that of adulterative Brucea javanica oil. Methods: The samples were determined by SSI HPLC and Alltech 2000 ESLD with a Waters C18 analytical column (200 mm×4.6 mm,5μm), byusingacetonitrile-dichloromethane (60 : 40) was used as the mobile phase at a flow rate of 1.2 ml·min^-1 and the detection temperature was 85℃, the column temperature was 40℃,and the injection volume was 15μl. Results: The fingerprint exhibited promising separation of each peak. The precision, reproducibility and stability of the method were good, and RSDs of the relative retention time and relative peak area were less than 3.0%, which were accordance with the technical re- quirements on the fingerprints of Chinese traditional medicines. On the basis of the method established by HPLC-ELSD, the fingerprint of Brucea javanica oil and that of adulterative Brucea javanica oil are established and the similarity is compared. Conclusion: This method provide important basis for the quality control and market supervision of Brucea javanica oil.
