摘 要:目的:研究弯柄灵芝多糖加速溶剂萃取技术(ASE)提取工艺。方法:以弯柄灵芝多糖含量为指标,蒽酮-浓硫酸法测定,并结合加速溶剂萃取法,采用L9(3^3)正交试验对弯柄灵芝多糖提取工艺优化设计。结果:加速溶剂萃取技术最佳提取工艺条件为提取温度180℃,提取时间12 min,静态循环提取次数2次,提取温度对其多糖提取率具有极显著性影响(P〈0.01)。在最优提取条件下,弯柄灵芝多糖提取率为(12.66±1.05)%。结论:该工艺与传统水提醇沉提取法相比,具有操作简便、缩短提取时间、节能耗、多糖得率高等优点,可用于弯柄灵芝多糖的提取生产,为弯柄灵芝产品的开发提供了有益的参考。Objective: To investigate a rapid extraction technology of polysaccharide from Ganodermaflexipes. Methods: The pol- ysaccharide content determined by an anthrone-sulfuric acid method was used as the index, an accelerated solvent extraction (ASE) was optimized by L9 (33 ) orthogonal experimental design to screen out the best extraction technique for G. flexipes. Results: The optimum ASE conditions were as follows: 180℃, twice static cycle extraction with 12 min per time. The extraction temperature was the most significant influencing factor. Under the optimal conditions, the yield of polysaccharide from G. flexipes reached as high as ( 12.66±1.05) %. Conclusion: Comparing with the traditional extraction technology of water extraction and alcohol precipitation, ASE technology is simple, rapid, economical and practical, which can be used for the extraction of polysaccharide from G. flexipes and provide beneficial reference for the products development.
