自发性乳腺癌小鼠的引进及其应用于澳洲茄碱抗癌药效评价

摘　　要：为了引进自发性乳腺癌小鼠模型并应用于澳洲茄碱在体抗癌药效评价，试验自国家遗传改造小鼠资源库引入SPF级MMTV—PyMT雄雌种鼠并进行培育，培育成功后以0．8，1．6，3．2mg／mL浓度的澳洲茄碱及其溶剂以按体重100肛L／10g腹腔注射MMTV-PyMT阳性小鼠，结合瘤块内加注100μL，每周一、周四各注射一次，连续注射6周，并以游标卡尺测量瘤块大小。结果表明：引进的MMTV—PyMT自发性乳腺癌小鼠模型能正常繁育并能再现其乳腺癌发病的经典病程，在药效评估上，与对照组相比，以0．8，1．6mg／mL浓度的澳洲茄碱腹腔注射100μL／10g结合瘤块内注射100μL，瘤块生长抑制率分别为14％-22％（P=0．06或P〈0．05）和38％-44％（P〈0．05），而腹腔注射3．2mg／mL组6周死亡率为87．5％（7／8）。说明MMTv—PyMT自发性乳腺癌小鼠模型引进成功，并且可应用于抗癌药物澳洲茄碱在体药效评价研究。To introduce a mouse model of spontaneous breast cancer and apply this model in the evaluation of the antieancer effect of Solasonine in vivo, both male and female SPF MMTV - PYMT mice introduced from the National Reseuree Center of Model Mice were euhivated in the test. Female MMTV - PyMT positive mice were treated with 100μL/10g solasonine and solvents at concentrations of 0.8, 1.6,3.2 mg/mL by intraperitoneal injection according to the body weight after successful cultivation or 100μL/10g intraperitoneal injection combined with 100μL in situ injection of tumor masses once on Monday and Thursday weekly. The injections continued for xix consecutive weeks, while the sizes of tumor masses were measured by the vernier caliper. The results showed that the introduced MMTV - PyMT mouse model of spontaneous breast cancer could normally reproduce and repeat the classical disease course of spontaneous breast cancer; in the evaluation of the drug effect, the tumor inhibitive rates were 14% to 22% （ P = 0.06 or P 〈 0.05 ） and 38 % to 44% （ P 〈 0.05 ） when using 100μL/10 g solasonine at concentrations of 0.8,1.6 mg/mL by intraperitoneal injection combined with 100 I.LL in situ injection of tumor masses compare with the control group, respectively, while the six - week mortality rate in the group treated with 3.2 mg/mL selasonine by intraperitoneal injection was 87.5 % （7/8）. The results indicate that the MMTV - PyMT mouse model of spontaneous breast cancer is successfully induced, and can be applied in the study on the evalntion of the anticancer efficacy of selsonine in vivo.
