摘  要:为了比较不同血清浓度的培养基对小鼠肺细胞原代培养的影响,在常规RPMI 1640培养基的基础上添加不同浓度的新生牛血清(5%、10%、20%、40%),每隔1 d在倒置显微镜下观察培养细胞的形态并用台盼蓝染色法统计细胞数目,绘制生长曲线。结果表明:4个血清浓度组中均有原代肺细胞生长增殖,血清浓度为10%的培养液培养的细胞生长状况最好,细胞形态完整清晰,数量较多,更适合小鼠的肺细胞生长。说明10%血清浓度的培养液最适合小鼠肺细胞的原代培养。To compare the effects of the culture media with different serum concentrations on primary culture of lung cells from mice, different concentrations of fetal calf serum were added to the RPMI 1640 culture media, including 5%, 10% ,20% and 40%, respectively. Cell morphol- ogy was observed under an inverted microscope every other day, meanwhile trypan blue staining wasused to record the number of cells, and then the cell growth curve was drawn. The results showed that the growth and proliferation of primary lung cells were found in the groups with 4 serum concentrations, and the growth of the cells cultured with the culture solution of 10% serum concentration was best; the cell morphology was complete and clear,a large number of cells were more suitable for the growth of lung ceils from mice. The results indicate that the culture solution of 10% serum concentrationis the most suitable concentration for primary culture of lung cells from mice.
