摘 要:目的:观察黄芪注射液对脑缺血再灌注大鼠海马Bcl-2、Bax、Caspase-3表达的影响。方法:雄性SD大鼠随机分为假手术组、脑缺血再灌注模型组、黄芪注射液溶剂组、黄芪注射液组,采用四血管阻断法建立脑缺血再灌注模型,分别采用免疫组化法和Western blot法检测海马Bcl-2、Bax、Caspase-3蛋白的表达。结果:与假手术组比,模型组Bcl-2、Bax、Caspase-3蛋白表达明显升高,Bcl-2/Bax明显降低(P<0.05);与模型组比,黄芪注射液组Bcl-2蛋白表达、Bcl-2/Bax明显升高,Bax、Caspase-3蛋白表达明显降低(P<0.05)。结论:黄芪注射液可通过升高Bcl-2的表达,降低Bax、Caspase-3的表达,抑制脑缺血再灌注大鼠海马神经元凋亡。Objective: To investigate the effects of astmgalus injection on Bcl-2, Bax and Caspase-3 expression in hiplx)campus of cerebral ischemia reperfusion rats. Methods: Male SD rats were randomly divided into sham-operation group, cerebral ischemia reperfusion model group, aslragalus injection solvent group and astragalus injection group. Four-vessel occlusion was used to establish cerebral ischemia reperfusion rats' model. was used to detect Bcl-2, Bax and Caspase-3 expression in hiippocampus. Results: Compared with sham-operation group, Bcl-2, Bax and Caspase-3 expression of rats in model group increased obviously, but Bcl-2/Bax decreased obviously (P 〈 0.05). Compared with model group, Bcl-2 and Bcl-2/Bax of rots in astragalus injection group increased obviously, but Bax and Caspased-3 decreased obviously (P〈 0.05). Condusions: Astragalus injection can inhibit neuronal apoptosis in of cerebral ischemia reperfusion rats by up regulating Bcl-2 expression and down regulating Bax, Caspase-3 expression.
