摘 要:目的观察硫化氢(H2S)对慢性应激抑郁模型大鼠行为和海马组织氧化应激的影响。方法雄性健康成年Wistar大鼠,随机分为正常对照组、应激抑郁(chronic unpredictable mild stress,CUMS)组、CUMS+低剂量NaHS(CUMS+LNaHS)组、CUMS+高剂量NaHS(CUMS+H-NaHS)组。采用CUMS加孤养复制大鼠抑郁模型。用强迫游泳实验检测大鼠行为学变化,用比色法检测海马组织中超氧化物歧化酶(SOD)的活性和丙二醛(MDA)的含量。结果与正常对照组比较,CUMS组大鼠强迫游泳不动时间显著延长,海马区SOD活性减低,MDA含量增高;与CUMS组比较,CUMS+L-NaHS组、CUMS+H-NaHS组大鼠强迫游泳不动时间均明显缩短,海马区SOD活性明显升高,MDA含量均明显下降。结论 H2S供体NaHS对慢性轻度不可预见性应激诱导的抑郁模型大鼠具有抗抑郁作用,其机制与提高大鼠的抗氧化应激能力有关。Objective To observe the effects of hydrogen sulfide (H2 S) on the behaviors ,SOD (superoxide dismutase ,SOD) activity and MDA (malonaldehyde ,MDA ) content of hippocampus in chronic stress depression rats .Methods Healthy male Wistar rats were randomly separated into normal control group ,CUMS group ,CUMS + L‐NaHS group ,CUMS + H‐NaHS group .A rat depression model was induced by CUMS .Behaviors of rats were assessed by forced swimming test and UV spec‐trophotometer analysis technique was used to detect SOD activity and MDA content of hippocampus .Results Immobility time of rat in forced swimming test increased ,SOD activity decreased and MDA content increased in the CUMS group rats .NaHS treatment could partly reverse these changes .Conclusion NaHS ,the donor of H2 S ,could relieve the depression induced by CUMS in rats ,and the mechanism may be correlated to increasing the antioxidative stress ability of rats .
