摘 要:低蛋白日粮往往引起肝脏IGF-I分泌降低,导致仔猪生长受限。为探讨其机制,本文采用代谢组学方法研究了肝脏关键代谢物对低蛋白的响应。试验选用体重接近的28日龄杜×长×大三元杂断奶仔猪18头,随机分为3组,每组6个重复。分别饲喂20%、17%和14%粗蛋白日粮并添加赖氨酸、蛋氨酸、色氨酸及苏氨酸达到NRC水平。预饲期5 d,正式试验30 d。结果表明:随着日粮蛋白水平的降低,血清中尿素氮、胰岛素、IGF-I含量,以及肝脏IGF-Im RNA表达水平亦随着降低。代谢组学研究表明,α-酮异戊酸和缬氨酸在14%CP日粮组显著降低;与此相反,血清总胆固醇、游离脂肪酸和肝脏IGFBP-1的m RNA表达水平却有所升高(P<0.05)。研究结果表明,低蛋白日粮造成仔猪生长受限与肝脏IGF-I和IGFBP-1的表达有关。肝脏缬氨酸和α-酮异戊酸等代谢关键物有可能在此过程中发挥重要作用。Low protein diets lead to the inhibition of liver IGF-I secretion and result in the growth restrictionofpiglets. To investigate the underlying mechanism, we screened the change of key metabolite inliver in response to low proteins by metabolomics methods. 28 days Duroc xLandrace xlarge piglets with similar average body weight were randomly divided into three groups (6 replicates per group) to fed with20%, 17% and 14% crude protein (CP) diet respectively. All the diets were supplemented with lysine, methionine, tryptophan and threonine to reach NRC level. After 30 days, animals were sacrificed to collect blood and liver samples. The results showed that with the decrease of dietary protein levels, serum urea nitrogen, insulin, IGF-I content, and IGF-I mRNA expression in the liver levels also decreased. Metabolomics studies showed14% CP group had the lowest level of ot-ketoisovalerate and valine significantly. In contrast, serum total cholesterol, free fatty acids and liver IGFBP-1 mRNA expression level were higher in 14% CP group than that in 20% CP group (P〈0.05). Our data confirmed that low-protein diets caused piglet growth restriction may attribute to the change of IGF-I and IGFBP expression inliver. Further, some key metabolites in liver, such asvaline and α-ketoisovalerate, are likely to play an important role in animal growth.
