强直性脊柱炎患者HLA-B27及基因亚型检测分析

摘　　要：目的通过对强直性脊柱炎患者(AS)HLA-B27阳性的标本进行基因亚型检测,了解各亚型的分布情况。方法应用流式细胞术检测HLA-B27抗原表达,荧光定量PCR检测基因亚型。结果正常人群中HLA-B27阳性率为7.1%(9/126),强直性脊柱炎患者HLA-B27阳性率为91.4%(53/58)。正常人群中基因亚型4种,分别为HLA-B2703、2704、2705、2706。强直性脊柱炎患者基因亚型3种,分别为HLA-B2702、2704、2705。结论 HLA-B27与强直性脊柱炎高度相关,基因亚型以B2704和B2705为主。Objective To investigate the distribution of HLA-B27 allele in patients with ankylosing spondylitis（AS）by detecting genetic subtypes.Methods The expression of HLA-B27 antigen was analyzed by flow cytometry.Genetic subtypes of HLAB27 was determined by fluorescence quantitative polymerase chain reaction assay.Results The positive rate of antigen HLA-B27 were 7.1%（9/126）and 91.4%（53/58）in healthy controls and AS patients respectively.There were four genetic subtypes in healthy controls：HLA-B2703,2704,2705 and 2706and three genetic subtypes in AS patients：HLA-B2702,2704 and 2705.Conclusion HLA-B27 is remarkably associated with AS.The prominent alleles are B2704 and B2705.
