摘 要:目的:探讨常用化痰类药物在博来霉素所致大鼠肺纤维化模型中的抗氧化作用,并对其抗氧化能力进行比较。方法:采用气管内滴入博来霉素(5mg/kg)的方法制作大鼠肺纤维化模型,以天南星、川贝母、海藻、胖大海、清半夏、白前、黄药子、前胡8味中药的水煎液及吡非尼酮对模型鼠进行灌胃干预28d,检测各组大鼠肺组织中SOD、GSH、MDA、Nrf2、Nrf2-mRNA表达水平,并对其抗氧化能力进行评分比较。结果:模型组SOD、GSH较空白组显著下降(P≤0.01),MDA明显升高(P≤0.01),Nrf2升高(P≤0.05);与模型组比较,吡非尼酮组、川贝母组、白前组、海藻组、前胡组均对SOD有明显改善作用(P≤0.01或P≤0.05),海藻组对GSH改善作用明显(P≤0.01),川贝母组、清半夏组、白前组MDA明显降低(P≤0.01),各给药组Nrf2表达均升高(P≤0.01)。综合评分结果显示川贝母、海藻的抗氧化能力较强。结论:化痰类中药对博来霉素所致的大鼠肺纤维化模型有明显的干预作用,其抗氧化作用以川贝母和海藻为突出,证实了化痰类中药可以通过干预机体的氧化/抗氧化机制对肺纤维化起到干预作用。Objective: To investigate and compare the antioxidant activities of common traditional Chinese medicines( TCMs) for resolving phlegm in rats with bleomycin- induced pulmonary fibrosis. Methods: A rat model of pulmonary fibrosis was induced by intratracheal instillation of bleomycin( 5 mg / kg). The model rats received water decoctions of Rhizoma Arisaematis,Fritillaria cirrhosa,algae,Sterculia lychnophora,Rhizoma Pinelliae,Cynanchum stauntonii,Dioscorea bulbifera,and Radix Peucedani and pirfenidone by gavage for 28 days. The levels of superoxide dismutase( SOD),glutathione( GSH),malondialdehyde( MDA),nuclear factor erythroid 2- related factor 2( Nrf2),and Nrf2- mRNA in the lung tissue of rats were measured,and the antioxidant capacity of these medicines was scored and compared. Results: Compared with the blank group,the model group showed significantly lower levels of SOD and GSH( P〈0. 01) and significantly higher levels of MDA( P〈0. 01) and Nrf2( P〈0. 05). Compared with the model group,the pirfenidone group,Fritillaria cirrhosa group,Cynanchum stauntonii group,algae group,and Radix Peucedani group had a significantly higher level of SOD( P〈0. 01 or P〈0. 05); the algae group had a significantly higher level of GSH( P〈0. 01); the Fritillaria cirrhosa group,Rhizoma Pinelliae group,and Cynanchum stauntonii group had a significantly lower level of MDA( P〈0. 01); every treatment group showed a significantly higher level of Nrf2( P〈0. 01). The composite score results showed that the antioxidant capacity of Fritillaria cirrhosa and algae was stronger. Conclusion: For rats with bleomycin- induced pulmonary fibrosis,the common TCMs for resolving phlegm have significant efficacy,and Fritillaria cirrhosa and algae have stronger antioxidant capacity. It suggests that the TCMs for resolving phlegm can exert the therapeutic effect on pulmonary fibrosis by regulating the oxidant / antioxidant system.
