摘 要:目的:观察制首乌汤剂对阿霉素大鼠血液生化指标和肾组织的影响,探讨其对肾病综合征的防治作用及机制。方法:SD大鼠28只随机分为空白组、模型组、益肾通络方组和制首乌组,每组7只,对除空白组外的其余组建立阿霉素肾病模型。制首乌组、益肾通络方组分别灌服制首乌汤剂(0.21g/m L)和益肾通络方汤剂(2.00g/m L)治疗,空白组和模型组灌服蒸馏水,每天1次,连续3周。采用双缩脲法检测造模前、灌药前及灌药后24h尿蛋白;灌药后第8周各组大鼠眼眶后静脉丛取血检测血液生化指标,摘取肾脏进行肾组织病理学检查。结果:灌药前模型组及给药组24h尿蛋白含量均>200mg,提示造模成功;灌药后给药组含量均降低(P<0.01或P<0.05),益肾通络方组改善优于制首乌组(P<0.05)。给药组生化指标均优于模型组(P<0.01),益肾通络方组较制首乌组改善更显著(P<0.05)。病理学检查结果显示给药组均对肾小球损伤有一定的修复。结论:制首乌单独用于治疗肾病综合征有一定效果,复方应用效果更显著。Objective: To observe the effects of Radix Polygoni Multiflori decoction on the blood biochemical parameters and renal histopathological changes in rats with adriamycin nephropathy,and to investigate its preventive and therapeutic effects on nephrotic syndrome and the mechanism. Methods: A total of 28 Sprague- Dawley( SD) rats were equally and randomly divided into four groups: blank group,model group,kidney- nourishing and collateral- dredging prescription group,and Radix Polygoni Multiflori group. An adriamycin nephropathy model was established in all groups except the blank group. The Radix Polygoni Multiflori group and the kidney- nourishing and collateral- dredging prescription group were given Radix Polygoni Multiflori decoction( 0. 21 g / m L) and kidney- nourishing and collateral- dredging prescription decoction( 2. 00 g / m L),respectively,by gavage. The blank group and the model group were given distilled water by gavage once every day for three weeks. The 24- h urinary protein excretion was determined by the biuret method before establishing the adriamycin nephropathy model and before and after administration. At 8 weeks after administration,the blood was collected from the posterior orbital venous plexus to measure the blood biochemical parameters,and the kidney was taken for histopathological examination. Results: Before administration,the 24- h urinary protein excretion was 〉200 mg in the model group,kidney- nourishing and collateral- dredging prescription group,and Radix Polygoni Multiflori group,which suggested that the model was established successfully. After administration,the kidney- nourishing and collateral- dredging prescription group and the Radix Polygoni Multiflori group showed a significant decrease in 24- h urinary protein excretion( P〈0. 01 or P〈0. 05),and the kidney- nourishing and collateral- dredging prescription group showed a significantly greater decrease in 24- h urinary protein excretion than the Radix Polygoni Multiflori group( P〈0. 05).
