益气养阴方对早期糖尿病肾病大鼠肾功能及MCP-1 mRNA表达的影响

摘　　要：目的:观察益气养阴方对早期糖尿病肾病大鼠的疗效并探讨其作用机制。方法:48只Wistar大鼠随机分为正常组、模型组、益气养阴方组、西药组。除正常组外,其余组均采用单侧肾脏切除、高脂高糖饮食联合STZ制备糖尿病肾病大鼠模型,观察各组大鼠治疗后血糖、尿白蛋白、血肌酐、尿素氮、单核细胞趋化蛋白-1(MCP-1)mRNA的变化。结果:模型组大鼠血糖、尿白蛋白较正常组高(P<0.05),益气养阴方组和西药组大鼠各项指标均好转,其中益气养阴方组血糖值及西药组血糖值、尿白蛋白值与模型组比较,差异有统计学意义(P<0.05);模型组肾脏组织内MCP-1 mRNA的表达较正常对照组高(P<0.05),益气养阴方干预后其表达下降(P<0.05)。结论:益气养阴方能减少糖尿病肾病大鼠血糖及尿白蛋白值,该作用可能与下调MCP-1 mRNA的表达有关。Objective： To investigate the clinical efficacy of prescription for reinforcing qi and nourishing yin in the treatment of rats with early diabetic nephropathy（ EDN） and its mechanism of action. Methods： A total of 48 Wistar rats were randomly divided into normal group,model group,prescription group,and Western medicine group. An EDN model was established in all the groups except for the normal group using unilateral nephrectomy and high- glucose and high-fat diet combined with streptozocin. The changes in blood glucose,urinary albumin,serum creatinine,urea nitrogen,and monocyte chemoattractant protein- 1 mRNA of each group after treatment were evaluated. Results： The model group had significantly higher blood glucose and urinary albumin levels than the normal group（ P〈0. 05）; all the indicators in the prescription and Western medicine groups were improved,and the blood glucose in the prescription group and the blood glucose and urinary albumin in the Western medicine group were significantly different from those in the model group（ P〈0. 05）; the model group had significantly higher expression of MCP- 1 mRNA in kidney tissue than the normal group（ P〈0. 05）,and the MCP- 1 mRNA expression was reduced after treatment with prescription for reinforcing qi and nourishing yin（ P〈0. 05）. Conclusion： The prescription for reinforcing qi and nourishing yin can reduce blood glucose and urinary albumin levels in rats with EDN,probably by down- regulating the MCP- 1 mRNA expression.
