摘 要:应用PCR技术检测江苏徐州地区三带喙库蚊Culex tritaeniorhynchus蚊胃血来源,研究其吸血习性。根据三带喙库蚊可能吸血对象(猪、人、牛、羊、鼠、鸡)的线粒体DNA细胞色素b序列的差异,设计种特异引物,建立聚合酶链反应(PCR)体系鉴定三带喙库蚊胃血来源。同时,用该方法检测猪血、人血、牛血、羊血、鼠血、鸡血中提取的DNA,验证其特异性;对吸饲人血后不同时间(1、12、24、36、48、60h)的中华按蚊进行检测,测试该方法的敏感性。该方法可从已知动物血样提取的DNA中分别扩增得到689(人)、561(牛)、453(猪)、225(羊)、485(鼠)、266bp(鸡)大小的特异性条带;吸人血36h内的中华按蚊均能扩增出特异性条带,在吸人血后48、60h的中华按蚊中,均未扩出特异性条带。共检测24只现场三带喙库蚊,血源来自猪、人、鸡、牛、鼠、羊分别为7、6、3,2、2、1只,其中5只蚊虫兼吸吸血。建立的PCR检测方法可鉴定三带喙库蚊蚊胃血来源,结果稳定可靠;徐州地区三带喙库蚊不仅嗜吸猪血和人血,也可兼吸其他动物血。To develop a PCR method for identifying the blood meal source and study the blo0d-sucking habit of Culex tritaeniorhynchus, the species-specific primers based on mitochondrial cytochrome b were used to identify the animal blood hosts of field-collected Cx. tritaeniorhynchus. Meanwhile, DNA was extracted from the hosts'blood to verify the specificity of the method. DNA extracted from Anopheles sinensis mosquitoes was analyzed to determine the sensitivity of the method !, 12, 24, 36, 48 and 60 h after ingesting a blood meal. Specific PCR products were amplified from the DNA extracted from different animal hosts. Host DNA was detectable in frozen mosquitoes for as long as 36 h after ingestion, but could not be amplified 48 h and 60 h after ingestion. PCR analysis of the DNA in wild-caught specimens indicated that Cx. tritaeniorhynchus fed mainly on the blood of pigs (29.2%) and human (25.0%). samples showed a mixed blood meal. The PCR assay was proved to be an accurate and reliable method of identifying the blood meals of wild-caught mosquitoes. Cx. tritaeniorhynchus in Xuzhou areas seemed to like to feed on pig and human as well as other animals.
