中药复方对ED大鼠海绵体Cx43的影响

摘　　要：目的基于"肝藏"理论,从阴茎海绵体平滑肌缝隙链接蛋白Cx43角度研究疏肝养血法对肝郁血虚ED大鼠的干预作用。方法选取经交配后证实有正常勃起功能的雄性SD大鼠30只,随机分为空白组、模型组、疏肝养血组,每组10只,采用尾部放血和悬吊激惹方法配合低铁饮食制备肝郁血虚勃起功能障碍模型,给以四物汤与逍遥散合方煎剂灌胃14天,末次灌胃24小时后处死大鼠,取阴茎海绵体组织,HE染色观察阴茎海绵体组织病理学变化,免疫组化SP法检测阴茎海绵体Cx43表达。结果与空白组相比,模型组阴茎海绵体组织病理学改变不明显,阴茎海绵体平滑肌中Cx43表达明显降低(P<0.05);与模型组相比,疏肝养血组阴茎海绵体平滑肌中Cx43表达显著提高(P<0.05)。结论改善阴茎海绵体平滑肌中Cx43表达可能是基于"肝藏"理论,疏肝养血法治疗ED的物质基础及作用机制之一。Objective To observe the effect of dispersing liver and nourishing blood treatment on Cx43 in penis smooth muscle of SD rats with the liver-stagnation and blood-deficiency erectile dysfunction based on the guidance of ＂Gancang＂ theory. Methods 30 normal erectile function sexually mature male SD rats which confirmed by mating with female rats were randomly divided into blank group, model group and dispersing liver and nourishing blood group, with 10 rats in each group, meanwhile they were fed with low-iron diet to establish the liver- stagnation and blood-deficiency ED models. After 14 days intragastrie administration with Siwu decoction and Xiaoyaosan decoction, all were killed after last gavage 24 hours with taking the sponge meristem to pathological observation with HE and the expression of Cx43 with SP. Results We found that the pathological changes of sponge meristem between blank group and model group were unobvious, but the expression of Cx43 in penis smooth muscle was declined significantly in the model group. Comparing with the model group, the expression of Cx43 in penis smooth muscle was increased in southing liver and nourishing blood group had statistical significance （ P 〈 0. 05 ）. Conclusions The dispersing liver and nourishing blood treatment based on the guidance of ＂Gancang＂ theory could effectively recovery the erectile functions through the mechanisms of affecting the expression of Cx43 in penis smooth muscle.
