艾塞那肽在高糖和低糖条件下对INS-1细胞胰腺十二指肠同源盒表达和细胞凋亡的影响

摘　　要：目的探讨胰高糖素样肽-1(GLP-1)类似物艾塞那肽在高糖和低糖条件下对胰岛B细胞株INS-1细胞胰腺十二指肠同源盒(PDX-1)表达和细胞凋亡的影响。方法体外培养INS-1细胞,分为正常对照组、高糖组、低糖组、高糖艾塞那肽干预组、低糖艾塞那肽干预组、高糖二甲双胍干预组。使用相差显微镜观察细胞生长状态,采用流式细胞仪双染法和原位TUNEL法检测INS-1细胞凋亡情况,分别采用实时PCR法、Western印迹法和免疫组织化学法检测PDX-1表达情况。结果高糖组和低糖组的细胞凋亡指数和细胞凋亡率均显著高于正常对照组(P值均<0.01),高糖组与低糖组间的差异均无统计学意义(P值均>0.05);高糖艾塞那肽干预组的细胞凋亡指数和细胞凋亡率均显著高于正常对照组(P值均<0.05),但均显著低于高糖组(P值均<0.05);低糖艾塞那肽干预组的细胞凋亡指数和细胞凋亡率均显著高于正常对照组(P值均<0.05),但均显著低于低糖组(P值均<0.05);高糖艾塞那肽干预组与低糖艾塞那肽干预组间细胞凋亡指数和细胞凋亡率的差异均无统计学意义(P值均>0.05);高糖二甲双胍干预组的细胞凋亡指数和细胞凋亡率均显著高于正常对照组(P值均<0.05),但与高糖组的差异均无统计学意义(P值均>0.05)。高糖组和低糖组的PDX-1表达均显著低于正常对照组(P值均<0.01),高糖组与低糖组间的差异无统计学意义(P>0.05);高糖艾塞那肽干预组的PDX-1表达显著低于正常对照组(P<0.05),但显著高于高糖组(P<0.05);低糖艾塞那肽干预组的PDX-1表达显著低于正常对照组(P<0.05),但显著高于低糖组(P<0.05);高糖艾塞那肽干预组与低糖艾塞那肽干预组间PDX-1表达的差异无统计学意义(P>0.05);高糖二甲双胍干预组的PDX-1表达显著低于正常对照组(P<0.05),但与高糖组的差异无统计学意义(P>0.05)。结论在高糖和低糖条件下,艾塞那肽均能抑制INS-1细胞的凋亡,同�Objective To observe the effect of Exenatide （a glucagon like peptide-1 analogue） on the expression of pancreatic duodenal homeobox-1 （PDX-1） and the apoptosis of islet beta-1 （INS-l） cells cultured with high and low glucose. Methods In vitro cultured INS-1 cells were divided into six groups： control group, high glucose （HG） group, low glucose （DG） group, high glucose ＋ Exenatide （HE） group, low glucose ＋ Exenatide （DE） group, and high glucose ＋ metformin （HM） group. Morphological changes of cells were observed under the inverted microscope. The apoptosis of INS-1 cells was tested by the flow cytometry and TdT-mediated dUTP nick end labeling. The real-time polymerase chain reaction （PCR）, western blot and immunohistochemical methods were used to detect the expression of PDX-1 in INS-1 cells. Results Both apoptosis rate and index of INS-1 cells in the HG and DG groups were significantly higher than those in the control group （all P〈0.01）, but no statistical difference was found between HG group and DG group （both P〉0.05）. The apoptosis rate and index in the HE group were significantly higher than those in the control group （both P〈0.05）, but significantly lower than those in the HG group （both P〈0.05）. The apoptosis rate and index in the DE group were significantly higher than those in the control group （both P〈0.05）, while significantly lower than those in the DG group （both P〈0.05）. There were no significant differences in the apoptosis rate or index between HE group and DE group （both P〉 0.05）. The apoptosis rate and index in the HM group were significantly higher than those in the control group （both P〈0.05）, while not statistically different from HG group （both P〉0.05）. PDX-1 expression in the HG and DG groups were significantly lower than those in the control group （both P〈0.01 ）, but no statistical difference was found between HG group and DG group （P〉0.05）.
