摘 要:目的探索母体绵羊妊娠期缺氧对胎羊心血管发育的影响及与心脏病理损伤的相关性。方法应用高原人工实验舱建立不同程度的妊娠期绵羊缺氧模型,分为平原对照组(LAC)、轻度低氧暴露组(MHE)和重度低氧暴露组(SHE)。应用超声检测胎羊的心血管发育情况,苏木素-伊红(HE)染色法及扫描电子显微镜检测评估胎羊心脏病理结构改变情况。结果超声检查未发现各组动物关键的心血管结构畸形存在。心脏组织样本HE染色及扫描电镜检测显示,MHE和SHE与LAC相比,实验动物的心脏组织出现了不同程度的细胞组织水肿,且线粒体分布异常。结论本实验初步证实高原环境下,母体妊娠期缺氧能够干扰胚胎的心血管系统生长发育,导致出生后心脏组织病理学结果发生异常。Objective To research the abnormal fetal cardiovascular development and cardiac pathological injury with the exposure of hypoxia environment in duration of pregnancy. Methods The different degrees of hypoxia pregnancy sheep mode by artificial cabin, and sixteen pregnancy sheep were divided into low altitude control group (LAC), mild hypoxia exposure group (MHE) and severe hypoxia exposure group (SHE). The development of sheep cardiovascular system was observed by using echocardiography. The sheep cardiac pathological changes was detected and evaluated by HE staining and scanning electron microscopy detection. Results The echocardiography results were showed that there was no significant change in the vital structure of heart for all groups. HE staining and electron microscopy scanning showed that the myocardium in MHE and SHE groups had different levels of tissue edema and abnormal distribution of mitochondrion compared to the LAC group. Conclusions This primary experimental results shows that the different degrees of hypoxia exposure cause negative impact on the development of pregnancy sheep fetal cardiovascular system, and influence the histopathology of heart after fetal birth.
