摘 要:目的分析人肠道病毒71型(EV-A71)感染神经母细胞瘤来源的SK-N-SH细胞的microRNA(miRNA)表达谱差异。方法用EV-A71病毒感染人SK-N-SH细胞,6h后收获总RNA,基因芯片检测感染和未感染EV-A71的神经细胞miRNA表达谱,并用实时荧光定量PCR方法 (qRT-PCR)进行验证。结果比较EV-A71感染与未感染的SK-N-SH细胞miRNA的表达谱,SK-N-SH细胞被EV-A71感染后有26种miRNA表达显著上调,40种miRNA表达显著下调。结论 EV-A71感染显著改变了SK-N-SH神经细胞的miRNA表达谱,可作为EV-A71感染机制深入研究的实验基础。Objective To analyze the expression profile of miRNA in neural cell line SK-N-SH, infected with human enterovirus 71 (EV-A71). Methods The total RNA of SK-N-SH neural cells infected with EVA71 for 6 hours was harvested, miRNA chip was used to detect the miRNA expression profile of cells, which was then verified by quantitative real-time PCR (qRT-PCR). Results A total of 26 miRNAs were up-regulated while 40 were down-regulated apparently in SK-N-SH cells infected with EV-A71 compared with non-infected cells. Conclusions The infection of EV-A71 can significantly change the miRNA expression profile of SK-N- SH cells, which may be used as the experimental basis for the further study of the mechanism of EV-A71 infection.
