摘  要:目的从JNK蛋白表达探讨清脑滴丸治疗急性脑缺血再灌注损伤引发细胞凋亡的作用机制。方法线栓法制备大鼠大脑中动脉缺血再灌注模型,随机分为假手术组、脑缺血1.5 h再灌注24 h模型组(I1.5hR24组)和清脑滴丸治疗组。TTC染色检测脑梗死面积,Western blot法检测大脑缺血侧皮层的JNK及磷酸化蛋白表达,TUNEL法检测细胞凋亡。结果与假手术组比较,I1.5hR24组大鼠脑梗死面积增大,p-JNK蛋白表达明显增加,细胞凋亡明显;清脑滴丸治疗组pJNK蛋白相对含量减少,细胞凋亡抑制,脑梗死面积减小,差异有统计学意义(P<0.01)。结论清脑滴丸可能通过抑制JNK磷酸化蛋白过表达,降低细胞凋亡,从而减轻脑缺血再灌注的损伤。Objective To discuss the action mechanism of Qingnao Dripping Pills in treating acute cerebral ischemia with reperfusion injury leading to apoptosis through JNK protein expression. Methods The rat models with cerebral middle artery ischemia with repeffusion were made by thread occlusion method. They were randomly divided into three groups: sham operation group, cerebral ischemia 1.5 h reperfusion 24 h group(I1. 5hR24 group), Qingnao Dripping Pills treatment group. TYC staining was used to measure cerebral infarction area, Western blot to detect the protein expression of JNK and p-JNK in the cortex of ischemia side, TUNEL method to measure the apoptosis. Results Compared with the sham group, the infarction area, the p-JNK protein expression in 11.5hR24 group were all increased and the apoptosis obviously seen. p-JNK protein expression was reduced, the apoptosis inhibited and the area of cerebral infarction lowered in Qingnao dripping pills group, the differenees were statistically significant( P 〈 0.01 ). Conclusion Qingnao Dripping Pills can probably inhibit p-JNK protein overexpression and decrease apoptosis, thereby reducing cerebral ischemia-reperfusion injury.
