自噬在镉致BRL 3A细胞损伤中的保护作用

摘　　要：以大鼠肝细胞系(BRL 3A)为模型,不同浓度的镉处理细胞,用免疫印迹法和免疫荧光法检测自噬标记分子LC3表达;通过添加雷帕霉素(自噬促进剂)和氯喹(自噬抑制剂)来升高和降低自噬水平,MTT法检测自噬水平的变化对细胞活性的影响。结果显示:与对照组相比,随着镉浓度的升高,细胞LC3-Ⅱ含量先上升后下降,8μmol/L时,LC3-Ⅱ含量最高,差异显著或极显著(P<0.05或P<0.01)。与镉处理组相比,自噬水平的上升可以显著提高细胞活性(P<0.05),自噬水平的下降显著降低细胞活性(P<0.05)。结果表明:镉可以引起BRL 3A细胞自噬水平的上升,激活的自噬对镉致细胞损伤具有一定的保护作用。To invest rat liver cell line （ associated protein igate the role of autophagy in cadmium-induced cytotoxicity in hepatocytes,the BRL 3A） was treated with different concentrations of cadmium. Microtubule- 1 light chain 3 （LC3）, a marker of autophagy, was examined by western blot and immunofluorescence staining. After rapamycin （an autophagic inducer） and chloroquine （an autophagic inhibitor） induce an increased and decreased level of autophagy,the effects of different levels of autophagy on cell viability were examined by MTT method. The results showed that compared with the control group,as the concentration of cadmium increased, the levels of LC3-Ⅱ proteins significantly increased and then decreased with the highest level of 8 μmol/L （P〈0.05 or P〈0.01）. Compared with the cadmium group, the increased level of autophagy could significantly improved the viability of cells （P〈0.05）, the decreased level of autophagy could reduced the viability of cells with significant difference （P〈0.05）. In summary,our results demonstrate that cadmium can induce autophagy, and autophagy serves an important protective role against cadmium-induced cytotoxicity.
