摘  要:[目的]观察大豆异黄酮对围绝经期综合征动物模型的影响,并探讨其作用机理,以期为临床上围绝经期综合征的治疗提供实验依据。[方法]选雌性SD大鼠50只随机分为5组,每组10只,分别为正常组、假手术组、模型组、乙烯雌酚组、大豆异黄酮组,除正常组和假手术组外,其余三组均摘除卵巢造模,形成围绝经期大鼠模型后,分别予生理盐水、乙烯雌酚、大豆异黄酮灌胃,正常组和假手术组分别予相同剂量生理盐水和大豆异黄酮灌胃。各组分别连续灌胃给药6周后,腹主动脉取血,分离血清,采用放射免疫分析法,测雌二醇(E2)、孕酮(P)、睾酮(T)水平。并分别摘取脑垂体、子宫、肾上腺组织,制备病理切片,用光镜观察实验效应,采用免疫组化法观察脑垂体中卵泡刺激素(FSH)和黄体生成素(LH)水平。[结果]放射免疫分析结果显示:大豆异黄酮组大鼠血清E2水平升高,与模型组比较有显著性差异(P<0.05),而对血清P、T水平影响不大。病理结果显示:大豆异黄酮组子宫壁及子宫内膜增厚介于模型组和假手术组之间,表面上皮呈柱状,腺体增多;大豆异黄酮组脑垂体中去势细胞介于模型组和正常组之间;大豆异黄酮组肾上腺组织改变不明显。免疫组化结果显示:大豆异黄酮组脑垂体组织中的FSH和LH细胞水平低于模型组。[结论]大豆异黄酮能使围绝经期综合征动物模型的子宫壁及内膜增厚,并具有提高其血清E2水平,但对血清P、T水平及肾上腺组织影响不大。[ Objective] To observe the effect of soybean isoflavone on the climacteric syndrome model of female rats, and to find its mechanism,in order to provide experimental evidence for the clinical treatment of menopausal syndrome. [ Methods ] 50 female SD rats were randomly divided into five groups, 10 rats in each group,including the normal control group,sham operation group,model group, diethylstilbestrol group, soybean isoflavone treated group, except the normal control group and sham operation group,the other groups were ovariectomized rats,the formation model of climacteric rats treated with normal saline,diethylstilbestrol ,soybean isoflavone by gavage,normal control group and sham operation group were given the same dose of normal saline and soybean isoflavone by gavage.After 6 weeks of continuous treatment, abdominal aorta was taken blood,the serum was isolated, and Radioimmunoassay method to measure Eestradiol (E2),Progesterone (P) ,Testosterone (T).And removal of the pituitary gland, uterine and adrenal prepared pathological section, experimental effect was observed by light microscopy, using method to observe the brain pituitary Follicle-Stimulating Hormone(FSH) and Luteinizing hormone(LH) levels. [ Results ] Radiation immune anal- ysis results show that soybean isoflavone can significantly improve serum E2 level in ovariectomized rats,compared with the model group, the statistical analysis shows significant difference(P〈0.05), but the serum P and T level have little effect. The pathological results show:Between the uterus: soybean isoflavone group of uterine wall and endometrial thickening between model group and sham operation group,the surface epithelium is columnar,glands increased. Between the pituitary gland: soybean isoflavone ovariectomized group cells between model group and normal group. Adrenal: soybean isoflavone group adrenal tissue did not change significantly. Immunohistocbemical results shown: Soybean isoflavone group pituitary tissues of FSH and LH cells
