大株红景天对百草枯中毒鼠肺组织NF-kβ表达的影响

摘　　要：目的:探讨大株红景天(Rosin)对百草枯中毒鼠肺核结合因子-kβ(NF-kβ)表达的影响。方法:120只SD雄性大鼠分为正常对照组(C)120只、百草枯中毒组(PQ)50只及大株红景天组(Rosin)50只。PQ组和Rosin组一次性灌胃PQ25mg/kg染毒,C组予等体积生理盐水灌胃。其中Rosin组于染毒前15min以Rosin 10mg/kg静脉注射保护,以后每日1次给药直至处死前;PQ组、C组分别在同时间点予与等体积生理盐水。观察各组大鼠在中毒后12h、1d、3d、5d、7d肺组织病理改变,检测肺泡灌洗液中NF-kβp65表达。结果:C组肺组织结构基本完整,PQ组炎性细胞浸润明显增多,肺组织损伤程度加重,Rosin组肺组织损伤程度较轻。PQ组NF-kβp65表达较C组显著增加,Rosin组降低。结论:NF-kβ在百草枯所致大鼠肺损伤中起重要作用,大株红景天降低肺组织NF-kβ水平,减轻百草枯中毒大鼠的肺组织损伤。Objective To investigate the expression of nuclear factor kappa β （ NF - kβ） in lung tissue of acute paraquat poi- soned rats and the protection of Rosin. Methods 120 male rats were randomly divided into 3 groups： normal control group （ C20 rats）, paraquat poisoning group （PQ50rats） and Rosin intervention group （Rosin 50rats）. The PQ and Rosin group rats were given paraquat by gastric perfusion （25mg/kg） ; The C group rats were given saline by gastric perfusion. The rats in Rosin group were pro- tected by intravenous injections of penis with Rosin （10mg/kg） 15min before exposure of paraquat, once a day until killed. The PQ and C group rats were given saline. Histological changes of lung tissues were observed at different time of 12h、1d、3d、5d and 7d after poisoned to determin expression of NF - kβP65. Results The degree lung injury in PQ group was higher than that in C group. But in Rosin group the degree of lung injury was lighter. Conclusions The content of NF - kβp65 in PQ group was higher than that in C group and that were lower in Rosin group. Conclusion NF - kβp65 could play an important role in lung injury of poisoned rats. Rosin may inhibit the expression of and NF - kβp65 and alleviate lung injury in acute paraquat poisoned rats.
