摘 要:目的:探讨人参皂苷Rg1对体外胚鼠皮层神经干细胞的增殖和神经胶质定向分化的影响。方法:本研究采用MTT法测定人参皂苷Rg1对胚鼠皮层神经干细胞的最佳剂量和最佳作用时间点,氧糖剥夺(OGD)后的细胞损伤模型,免疫荧光染色鉴定神经干细胞增殖和神经胶质样定向分化。结果:分离后体外培养的胚鼠皮层细胞表达Nestin并自我复制,为神经干细胞,人参皂苷Rg1对神经干细胞增殖的最佳剂量是0.016μg/m L,0.016μg/m L人参皂苷Rg1对神经干细胞的增殖最佳作用时间点是6h。在人参皂苷Rg1的作用下,神经干细胞及OGD后的神经干细胞中Nestin、Brd U、光密度、阳性细胞Vimentin的面密度数目及Nestin/Brd U、Nestin/Vimentin双标染色细胞数目有显著提高(P<0.05,P<0.01)。结论:人参皂苷Rg1具有促进皮层神经干细胞增殖和神经胶质样定向分化的作用,这一机制可能是人参皂苷Rg1中具有类似于生长因子或刺激某些细胞分泌生长因子,促进神经干细胞的胶质样分化和增殖。Objective: To investigate the effects of Ginsenoside-Rg1 on proliferation and neuroglia directed differentiation of cortical neural stem cells(NSCs) of embryonic rat in vitro. Methods: This study used MTT method to screen the best effective dose and window time of Ginsenoside-Rg1 on proliferation of cortical neural stem cells of embryonic rat, and oxygen glucose deprivation(OGD) for cell injury model. Immunofluorescence staining was used for identification of proliferation and neuroglia directed differentiation of NSCs. Results: The isolated cortical cells of embryonic rat were cultivated in vitro and expressed Nestin with self-duplication, which were NSCs. The best effective dose of Ginsenoside-Rg1 on proliferation of NSCs is 0.016μg/mL. The best effective window time of 0.016μg/mL Ginsenoside-Rg1 on proliferation of NSCs is 6h. GinsenosideRg1 promotes expression of Brd U, Nestin, and Vimentin in primary and passage NSCs and NSCs after OGD(P〈0.05, P〈0.01). Conclusion: Ginsenoside-Rg1 has a role in promoting proliferation and neuroglia directed differentiation of cortical neural stem cells. The possible reason would be that Ginsenoside-Rg1 has the effects similar to the growth factors or stimulating some cells to secrete the growth factors to promote the proliferation and differentiation of NSCs.
