摘  要:目的:研究不同浓度片仔癀对人骨肉瘤MG63细胞增殖的影响以及PI3K/Akt信号通路的作用机制。方法:1不同浓度片仔癀作用于骨肉瘤MG63细胞,用MTT法检测其对MG63细胞增殖的抑制作用。2流式细胞仪检测其作用于MG63细胞后细胞的周期变化。3Western Blot法检测MG63细胞增殖过程中PI3K/Akt信号通路的关键蛋白PI3K、Akt、P-Akt的含量。结果:片仔癀能够抑制MG63细胞的增殖,呈剂量依赖性。片仔癀能使细胞在G2期的阻滞增加,降低PI3K/Akt信号通路中关键蛋白PI3K、Akt、P-Akt的表达,有剂量依赖性。结论:片仔癀可以通过降低PIK/Akt信号通路中关键蛋白PI3K、Akt、P-Akt的表达,有效抑制骨肉瘤MG63细胞的增殖。Objective: To study the effects of different concentrations of Pien Tze Huang on human osteosarcoma MG63 cell proliferation and mechanism of PI3K/Akt signal pathway. Methods: 1 After intervention of different concentrations of Pien Tze Huang on osteosarcoma MG63 cells, its inhibitory effect is detected by MTT method. 2Cell cycle of MG63 cells is detected by flow cytometry. 3 The key protein PI3 K, Akt and P-Akt of PI3K/Akt signal pathway of MG63 cell proliferation are detected by Western Blot. Results: Pien Tze Huang can inhibit the proliferation of MG63 cells, and there exists a time-dose dependent. Pien Tze Huang can inhibit the G2 phase cells, reduce the expression of PI3 K, Akt and P-Akt of PI3K/Akt signaling pathway, and there is a dosage dependent relationship. Conclusion: Pien Tze Huang can reduce the expression of PI3 K, Akt and P-Akt of PIK/Akt signaling pathway, and inhibit osteosarcoma MG63 cell proliferation effectively.
