木香挥发油对斑马鱼胚胎发育毒性的初步研究

摘　　要：目的:探讨梯度浓度木香挥发油对斑马鱼胚胎发育的影响。方法:用梯度浓度的木香挥发油处理2、12、24hpf的斑马鱼胚胎,36h后检测木香挥发油对不同发育期斑马鱼的半数致死剂量(LD50),同时分别在加药后12、24、36h记录木香挥发油对12hpf或24hpf斑马鱼胚胎畸形率以及心脏形态和心率的影响。结果:药物作用36h后,木香挥发油对2、12、24hpf斑马鱼胚胎的LD50分别为2.35、6.83、26.9μg/m L;在浓度>3μg/m L时,木香挥发油对12hpf的斑马鱼胚胎表现出明显的致畸性,对24hpf的斑马鱼表现出延迟发育和一定的特异性心脏毒性,导致心脏畸形、心包水肿、心率随浓度增大而减小。结论:木香挥发油对斑马鱼胚胎发育有一定毒性,而且发育期越早,毒性越大,对24hpf的斑马鱼表现为延迟发育和特定的心脏毒性。Objective： To investigate the influence of the gradient concentration volatile oil of common aucklandia root on zebrafish embryos. Methods： Zebrafish embryos of 2, 12, 24 hours post fertilization（hpf） were treated with the gradient concentration volatile oil. Lethal doses 50（LD50） of zebrafish at different developmental stages was detected 36 h after treatment, meanwhile the cardiac phenotypes, heart rates and embryo malformation rate of zebrafish were recorded respectively at 12, 24, 36 h after treatment. Results： The LD50 of zebrafish embryos of 2, 12, 24 hpf were 2.35, 6.83 and 26.9μg/mL respectively 36 h after treatment. When the concentration of volatile oil was higher than 3μg/mL, significant teratogenicity was found on 12 hpf zebrafish embryos, while retardation and certain specific cardiac toxicity such as heart malformation, pericardial edema and heart rate decreased with the increasing concentration were discovered on 24 hpf zebrafish. Conclusion： Volatile oil of Common aucklandia root showed certain toxic effects on early developmental stage of zebrafish embryo, while retardation and certain specific cardiac toxicity were founded on 24 hpf zebrafish.
