摘 要:目的:研究大孔树脂纯化七叶莲总三萜的工艺条件及参数。方法:采用紫外分光光度法测定七叶莲总三萜含量,选择3种大孔树脂,以总三萜吸附率和解吸率为指标,筛选适宜树脂类型,并对分离纯化工艺进行优化。结果:AB-8树脂对总三萜的吸附解吸效果最好。最佳纯化工艺条件:以自然p H条件下的样液上样,上样浓度为0.884mg/m L,最大上样量6BV,用8BV的80%乙醇进行洗脱。该工艺下产品的洗脱率达48.14%,纯化后总三萜的质量分数达45.98%。结论:AB-8大孔树脂分离纯化七叶莲总三萜工艺效果好,简单可行,可为工业化纯化七叶莲总三萜提供理论依据。Objective: To study the purification process of total triterpenoids from Schefflera arboricola Hayata by macroporous resin. Methods: Firstly, the content of total triterpenoids was analyzed by ultraviolet spectrophotometry. Then, the most optimal macroporous resin was selected from three kinds of resins on the basis of the adsorption rate and desorption rate of total triterpenoids. Finally, the main process conditions were ascertained to optimize the process parameters. Results: AB-8 type resin had the best adsorption and desorption effects among the three kinds of resins. The optimal process conditions were as following: the concentration of sample at natural p H level was 0.884mg/mL, the maximum loading amount was 6BV, and solvent of elution was 80% of alcohol(8BV). Under the conditions, the desorption rate reached 48.14% and the content of total triterpeniods in extract was 45.98%. Conclusion: It is effective, simple and feasible that AB-8 macroporous resin is used to extract the total triterpenoids from Schefflera arboricola Hayata, which will provide a theoretical reference for industrial purificaiton.
