黄芪注射液对糖尿病心肌病大鼠左心室功能及血清GLP-1水平的影响

摘　　要：目的观察黄芪注射液对糖尿病心肌病大鼠左心室功能及血清胰高血糖素样肽-1(GLP-1)水平的影响。方法 Wister大鼠随机分为对照组和糖尿病组,将成模糖尿病大鼠随机分为心肌功能正常组与心肌病组两组,心肌病组给予阿霉素腹腔注射。采用超声仪检测大鼠左心室整体功能,采用左室射血分数(LVEF)<42%作为糖尿病大鼠心肌病模型造模成功的标准。将成模心肌病组大鼠随机分为模型对照组与黄芪组,黄芪组每日将60 mg/m L黄芪注射液按200 mg/kg灌胃给药。心肌功能正常组为对照组。观察黄芪对大鼠左心室整体功能的影响及血清GLP-1水平。结果与对照组相比,模型对照组及黄芪组LVEF、左室短轴缩短率(LVFS)、GLP-1水平明显下降(P<0.05)。与模型对照组相比,黄芪组LVEF、左室短轴缩短率(LVFS)、GLP-1水平明显升高,比较有显著差异(P<0.05)。GLP-1水平与LVEF、LVFS呈正相关(相关系数为r=0.35,P<0.05;r=0.26,P<0.05)。结论黄芪注射液可以改善2型糖尿病心肌病大鼠的心室功能及提高2型糖尿病心肌病大鼠血清GLP-1水平,GLP-1水平与LVEF、LVFS呈正相关。Objective： To explore the effect of astragalus injection on the left ventricular function and plasma levels of Glp-1 in rats with type 2 diabetic cardiomyopathy. Methods： Wister rats were randomly divided into the control group and the diabetic group. The diabetic rats were randomly divided into 2 groups： the normal myocardial function group and the cardiomyopathy group. The cardiomyopathy group was given intraperitoneal injection of adriamycin. We used ultrasonic instrument to detect the left ventricular global function in rats and took EF〈 42% as the success criteria on building the rats model with type 2 diabetic cardiomyopathy. The rats with type 2 diabetic cardiomyopathy were randomly divided into the model control group and the astragalus group. The astra- galus group was infused with 60 mg/mL astragalus injection per day at dose of 200 mg/kg body mass. The normal myocardial function group was considered the control group. The effect of astragalus on the left ventricular func- tion and plasma levels of Glp-1 in rats with type ：2 diabetic cardiomyopathy was explored. Results： Compared with The control group ,the LVEF, LVFS, glp-1 levels of the model control group and the astragalus group significantly decreased （P〈 0.05 ）. Compared with the model control group ,the LVEF,LVFS, glp-1 levels of astragalus group significantly elevated （P〈 0.05）. Glp-1 levels and LVEF, LVFS were positively correlated （correlation coef- ficient of r = 0.35, P 〈 0.05 ; r = 0.26, P 〈 0.05）. Conclusion： Astragalus injection can improve the ventricular func- tion and the serum levels of glp-1 of type 2 diabetic cardiomyopathy rats. Glp-1 levels and LVEF, LVFS are pos- itively correlated.
