摘 要:目的:研究川芎水提物对缺氧人脐静脉内皮细胞(EAHY926)的作用影响。方法:体外培养EAHY926,采用化学诱导缺氧建立模型,用川芎水提取液干预后,测定细胞活力,并检测丙二醛含量、超氧化物歧化酶及一氧化氮的变化。结果:采用单因素方差分析,与模型组相比,川芎水提取液高剂量组、中剂量组及复方丹参组均能显著提高细胞活力(P<0.05);川芎水提物中剂量组、高剂量组及复方丹参组丙二醛含量与模型组比较有显著差异(P<0.01);川芎水提物高剂量组及复方丹参组一氧化氮含量显著增加(P<0.01);川芎水提物中剂量组及复方丹参组超氧化物歧化酶活性显著升高(P<0.05),川芎水提物高剂量组效果最好(P<0.01)。结论:川芎水提物能显著拮抗缺氧对EAHY926的损伤。Objective: To study the pharmacological effects of Ligusticum Chuanxiong water extract on hypoxia of human umbilical vein endothelial cells( EAHY926). Methods: In vitro culture of EAHY926 was performed by chemical hypoxia to establish model.After intervention by Ligusticum Chuanxiong water extract,we determined the cell viability,and detected the changes of malondialdehyde( MDA),superoxide dismutase( SOD) and nitric oxide( NO). Results: Data colleted was treated by the single factor variance analysis. Compared with the model group,water extract of Ligusticum Chuanxiong liquid high,middle dose group and Salvia miltiorrhiza group can significantly improve the cell viability( P〈0. 05); High,middle dose group of Ligusticum Chuanxiong and compound salvia miltiorrhiza group decreased significantly of MDA( P〈0. 01); High,middle dose group of Ligusticum Chuanxiong and compound salvia miltiorrhiza group increased significantly of NO content( P〈0. 01); Middle dose group of Ligusticum Chuanxiong and compound salvia miltiorrhiza group significantly increased SOD activity( P〈0. 05),and high dose group of Ligusticum Chuanxiong effect made the best( P〈0. 01). Conclusion: The water extract of Ligusticum Chuanxiong is significantly effective in antagonizing the hypoxia injury on EAHY926.
