保肝解毒颗粒对急性肝损伤小鼠肝细胞Bcl-2、Bax表达的影响

摘　　要：目的:观察保肝解毒颗粒对雷公藤多苷所致小鼠急性肝损伤肝细胞凋亡相关基因Bcl-2、Bax表达的影响,探讨保肝解毒颗粒的干预作用。方法:60只昆明种清洁级小鼠随机分为6组,即空白组,模型组,对照组,保肝解毒颗粒给药高剂量组、中剂量组、低剂量组。造模前,每天1次,给予小鼠灌胃不同剂量的保肝解毒颗粒,连续5 d;对照组灌服甘利欣混悬液;空白组、模型组灌服生理盐水。第5天给药后禁食12 h,以雷公藤多苷造模。免疫组织化学法检测肝细胞Bcl-2、Bax基因表达。结果:与正常组对比,模型组Bcl-2蛋白表达下降(P<0.05),而Bax蛋白阳性表达升高(P<0.05);保肝解毒颗粒组与模型组对比,下调Bax蛋白表达(P<0.05),上调Bcl-2蛋白表达(P<0.05)。结论:保肝解毒颗粒可以下调促凋亡基因Bax水平,上调抗凋亡基因Bcl-2水平,抑制细胞凋亡,从而起到对肝脏的保护作用,这可能是其治疗急性肝损伤的机制之一。Objective： To observe the effect of Baogan Jiedu Granule on liver cell apoptosis related gene expression of Bcl-2,Bax changes caused by the tripterygium glycosides inducing acute liver injury in mice,and to explore its intervention effect of protecting liver. Methods： 60 mice of Kunming-strain were randomly divided into untreated group,positive group,model group,low,medium and high doses groups of Baogan Jiedu Granule. Before establishing models,low,medium and high doses groups were respectively perfused with the different dosage of Baogan Jiedu Granule for successive 5 days. Positive group was administered by Ganlixin capsule. Simultaneously,untreated group and model group were given equivoluminal distilled water. Then all groups were perfused once to make model with tripterygium glycosides except for control model. After 12 hours. Bcl-2 and Bax were observed by using the method of immunohistochemical method. Results： Compared with control group,the gene expression of Bcl-2 in the serum of model group reduced obviously（ P〈0. 05）. Compared with model group,the gene expression of Bcl-2 were all improved in each dosage of groups,especially in medium-dosage（ P〈0. 05）; compared with control group,the gene expression of Bax in the serum of model group increased obviously（ P〈0. 05）. Compared with model group,the gene expression of Bax could be degraded in each dosage of groups（ P〈0. 05）. Conclusion： Baogan Jiedu Granule can repress apoptosis of the liver cells,effectively reduce the levelof Bax,increase Bcl-2,which can play a role to protect liver,and it may be one of the mechanism of the treatment of acute liver damage.
