兔动脉粥样硬化心脏局部RAS及TIMP-4对心脏胶原的影响及阿托伐他汀的干预

摘  要:目的建立兔动脉粥样硬化模型,观察兔动脉粥样硬化时心脏局部糜酶和TIMP-4的表达对心脏胶原的影响以及阿托伐他汀的干预效果。方法18只雄性新西兰大白兔,随机分为3组:正常对照组、动脉粥样硬化模型组、阿托伐他汀组。检测心脏局部糜酶及TIMP-4的表达、心脏局部AngⅡ含量及心脏胶原含量。结果AS组心脏糜酶及心脏局部TIMP-4的表达显著高于C组和ATV组(P〈0.01);AS组的心脏局部AngⅡ及心脏胶原含量显著高于C组和ATV组(P〈0.01)。结论动脉粥样硬化时心脏局部RAS的激活主要表现在糜酶、TIMP-4的表达增强和AngⅡ的明显升高,引起心肌胶原增生和纤维化。阿托伐他汀可以通过抑制糜酶及TIMP-4的表达来抑制心脏局部RAS的激活,从而抑制心肌胶原增生及纤维化。Objective To establish rabbit atherosclerosis model and to observe the change of cardiac TIMP-4, investigate the chang of cardiac collagen and the effect of atorvastatin on the expression of cardiac collagen in atherosclerotic rabbits. Methods Eighteen male New Zealand white rabbits were randomly divided into three groups: control group, atherosclerosis group, atorvastatin group. To determine the expression of cardiac Chymase and TIMP-4, the level of cadiac Ang Ⅱ and cardiac collagen. Result The cardiac chymase expression and cardiacTIMP-4 of AS Group higher than C Group and ATV group(P〈0.01) ;The levels of cardiac Ang Ⅱ and cardiac collagen in AS group were significantly higher than the C group and ATV group(P〈0.01). Conclusions When atherosclerosis occurs, the activation of cardiac reninangiotensin system mainly in the expression and level of chymase,TIMP-4,Ang Ⅱ and cardiac coUagen were markedly elevated, which cause collagen hyperplasia and fibrosis of myocardium. Atorvastatin could restrain cardiac renin-angiotensin system, and prevent the hyperplasia of cardiac collagen and the fibrosis of cardiac extracellular matrix.
